TOP STORYResearchers demonstrated that the clinically validated protein phosphatase 2A (PP2A) inhibitors LB100 and LB102, in combination with tyrosine kinase inhibitors (TKIs), selectively eliminate treatment-naïve TKI-insensitive stem and progenitor cells, while sparing healthy counterparts. PUBLICATIONS(Ranked by impact factor of the journal)Investigators found that connexin 26 (Cx26) was elevated in self-renewing CSCs and was necessary and sufficient for their maintenance. Cx26 promoted CSC self-renewal by forming a signaling complex with the pluripotency transcription factor NANOG and focal adhesion kinase (FAK), resulting in NANOG stabilization and FAK activation. [Nat Commun] Full Article | Press Release A model of chronic myeloid leukemia (CML) stem cells derived from CML induced pluripotent stem cells identified ADAM8 as an antigen of tyrosine kinase inhibitor-resistant CML cells. [Stem Cell Reports] Full Article Scientists investigated the role of chromobox protein homolog 7 (CBX7) in regulating stem cell-like properties of gastric cancer and the underlying mechanisms. [J Hematol Oncol] Full Article IKKα Is Required in the Intestinal Epithelial Cells for Tumor Stemness The authors generated a compound mouse model with mutation in Adenomatous Polyposis Coli and lacking intestinal epithelial IKKα, produced intestinal organoids and tumor spheroids with different genetic backgrounds, and performed immunohistochemistry and RNA-seq analysis. [Br J Cancer] Abstract Researchers previously demonstrated that BRM270 suppresses tumorigenesis by negatively regulating nuclear factor-κB signaling in multidrug-resistant cancer stem cells. In this study they report that the growth, migration, and invasion of normal human lung adenocarcinoma cells and their chemoresistant derivatives was inhibited by BRM270 treatment. [Cell Death Dis] Full Article PDE5 Inhibition Eliminates Cancer Stem Cells via Induction of PKA Signaling To identify novel CSC targeting compounds, scientists set-up an aldehyde dehydrogenase-based phenotypic screening system that allows for an automated and standardized identification of CSCs. [Cell Death Dis] Full Article Using global expression analysis of miRNAs in 10 paired in vitro as well as in vivo characterized primary glioblastoma stem-like cell and non-stem glioblastoma cultures, the authors identified a miRNA signature associated with the stem-like phenotype in glioblastoma. [Sci Rep] Full Article Investigators found that Myc-associated zinc-finger protein (MAZ) is predominantly expressed in pancreatic cancer stem cells. Functional analysis indicated that MAZ depletion in pancreatic ductal adenocarcinoma (PDAC) cells inhibits invasive phenotypes such as the epithelial-to-mesenchymal transition, migration, invasion, and the sphere-forming ability of PDAC cells. [J Biol Chem] Abstract | Full Article Organoids with Cancer Stem Cell-Like Properties Secrete Exosomes and HSP90 in a 3D Nanoenvironment The authors examined morphologies of 67 cell lines cultured on three dimensional morphology enhancing nanoculture plates and classified the types of cellular aggregates that form. Among the 67 cell lines, 49 cell lines formed spheres or spheroids, 8 cell lines formed grape-like aggregation, 8 cell lines formed other types of aggregation, and 3 cell lines formed monolayer sheets. [PLoS One] Full Article Cellular Toxicity Driven by High-Dose Vitamin C on Normal and Cancer Stem Cells Scientists identified that high-dose vitamin C shows cellular toxicity on proliferating NSPCs. They also demonstrated that undifferentiated NSPCs are more sensitive to vitamin C-driven DNA damage than differentiated cells, due to higher expression of Glut genes. POLICY NEWSTrump Science Budget Sows Confusion US president makes last-minute decision to abandon proposal for major cuts to National Institutes of Health, National Science Foundation and Department of Energy’s science office. [Nature News] Editorial Researchers Debate Whether Journals Should Publish Signed Peer Reviews Scientific journals should start routinely publishing the text of peer reviews for each paper they accept, said attendees at a meeting last week of scientists, academic publishers, and funding organizations. But there was little consensus on whether reviewers should have to publicly sign their critiques, which traditionally are accessible only to editors and authors. [ScienceInsider] Editorial
