PUBLICATIONS(Ranked by impact factor of the journal)Researchers report that extracellular matrix through laminin-integrin α6 upregulated ten-eleven translocation enzyme 3 (TET3) dioxygenase. TET3 in turn mediated DNA cytosine 5′-hydroxymethylation and upregulated genes critical for maintenance of glioma stem cells. [Oncogene] Abstract Biological function analyses, cell half maximal inhibitory concentration, self-renewal, and migration and invasion capacities, were detected by CCK8, sphere formation and Transwell assays. [J Exp Clin Cancer Res] Full Article Scientists addressed the effects of, and interactions between, ecotropic virus integration site 1 (EVI1) and retinoids in acute myeloid leukemia (AML) leukemic stem cells. They report that Evi1 reduced the maturation of leukemic cells and promoted the abundance, quiescence, and activity of leukemic stem cells in an MLL-AF9-driven mouse model of AML. [Cell Death Dis] Full Article Researchers identified critical autocrine factors mediating glioma-initiating cell survival and evaluated the anti-glioblastoma (GBM) effect of antagonizing these factors. Proteomic analysis was performed using conditioned media from two different patient-derived glioblastoma tumor spheres under a growth factor-depleted status. [Exp Mol Med] Full Article Mechanistically, treatment with iRGD-polymersome-tamoxifen resulted in inhibition estrogen receptor transcriptional activity as evaluated by a luciferase reporter assay. iRGD-polymersome-tamoxifen reduced the number of cells with self-renewing capacity, a characteristic of breast cancer stem cells. [J Nanobiotechnology] Full Article Overexpression of miR-199a-5p reduced the stemness of cutaneous squamous cell carcinomas and inhibited cell proliferation. By targeting the deacetylase Sirt1, miR-199a-5p inhibited cellular proteolysis of CD44 and reduced CD44 intracellular domain (CD44ICD) release and nuclear translocation. [Cell Cycle] Abstract Investigators showed evidence that Ku80 physically interacted with SALL4. The interaction competitively disrupted the SALL4-OCT4 complex and resulted in OCT4 lysosomal degradation. Ku80 inhibited self-renewal and metastasis of hepatocellular carcinoma cells through breaking the SALL4-OCT4 interactions and down-regulating the expression of OCT4. [Int J Biochem Cell Biol] Full Article The authors report the synthesis and biological evaluation of small molecules with chloroacetamide war-head. Their optimism yielded four hits inhibiting self-renewal of cancer cells with stem-like characters in vitro. INDUSTRY NEWSThe LeukoStrat® CDx FLT3 Mutation Assay may now be used as an aid in the assessment of AML patients for treatment with XOSPATA® in Europe. FLT3 mutation must be confirmed with a validated test, such as the LeukoStrat CDx FLT3 Mutation Assay. [Invivoscribe, Inc. (GlobeNewswire, Inc.)] Press Release Cyclacel Pharmaceuticals, Inc. announced study design and preliminary data from two of the Company’s Phase I studies evaluating a combination of CYC065, a CDK2/9 inhibitor, with venetoclax, a BCL2 inhibitor, to treat patients with relapsed or refractory acute myeloid leukemia or myelodysplastic syndromes and chronic lymphocytic leukemia. POLICY NEWSKU Leuven Investigates Whether Stem Cell Scientist Falsified Data KU Leuven is investigating allegations that one of its researchers, acclaimed stem cell biologist Catherine Verfaillie, was involved in falsifying research in at least 10 scientific papers published between 1999 and 2018, Belgian newspaper De Tijd reported last week. [The Scientist] Editorial Congress Set to Form Groups to Protect US Research Security Congress is expected to approve an annual defense bill as early as this week that would create two federal bodies aimed at preventing foreign government influence in scientific research. [The Scientist] Editorial US Scientists Who Hide Foreign Ties Should Face Research Misconduct Sanctions, Panel Says US scientists who violate government rules on disclosing foreign research ties should be investigated for research misconduct, says an independent group of prominent scientists asked to examine the threat of foreign influences on the US research enterprise.
