Integrin α6 Signaling Induces STAT3-TET3-Mediated Hydroxymethylation of Genes Critical for Maintenance of Glioma Stem Cells

Researchers report that extracellular matrix through laminin-integrin α6 upregulated ten-eleven translocation enzyme 3 (TET3) dioxygenase. TET3 in turn mediated DNA cytosine 5′-hydroxymethylation and upregulated genes critical for maintenance of glioma stem cells. [Oncogene] Abstract

Scientists addressed the effects of, and interactions between, ecotropic virus integration site 1 (EVI1) and retinoids in acute myeloid leukemia (AML) leukemic stem cells. Secretome Analysis of Patient-Derived GBM Tumor Spheres Identifies Midkine as a Potent Therapeutic Target

Researchers identified critical autocrine factors mediating glioma-initiating cell survival and evaluated the anti-glioblastoma (GBM) effect of antagonizing these factors. iRGD-Guided Tamoxifen Polymersomes Inhibit Estrogen Receptor Transcriptional Activity and Decrease the Number of Breast Cancer Cells with Self-Renewing Capacity

Mechanistically, treatment with iRGD-polymersome-tamoxifen resulted in inhibition estrogen receptor transcriptional activity as evaluated by a luciferase reporter assay. iRGD-polymersome-tamoxifen reduced the number of cells with self-renewing capacity, a characteristic of breast cancer stem cells. miR-199a-5p Represses the Stemness of Cutaneous Squamous Cell Carcinoma Stem Cells by Targeting Sirt1 and CD44ICD Cleavage Signaling

Overexpression of miR-199a-5p reduced the stemness of cutaneous squamous cell carcinomas and inhibited cell proliferation. By targeting the deacetylase Sirt1, miR-199a-5p inhibited cellular proteolysis of CD44 and reduced CD44 intracellular domain (CD44ICD) release and nuclear translocation. [Cell Cycle] Abstract

Ku80 Negatively Regulates the Expression of OCT4 via Competitive Binding to SALL4 and Promoting Lysosomal Degradation of OCT4

Investigators showed evidence that Ku80 physically interacted with SALL4. The interaction competitively disrupted the SALL4-OCT4 complex and resulted in OCT4 lysosomal degradation. Ku80 inhibited self-renewal and metastasis of hepatocellular carcinoma cells through breaking the SALL4-OCT4 interactions and down-regulating the expression of OCT4.
