    Generation and Genetic Engineering of Human Induced Pluripotent Stem Cells Using Designed Zinc Finger Nucleases
    Zinc finger nuclease-mediated gene targeting was used to generate both single allele CCR5-modified human induced pluripotent stem cells (hiPSCs) and biallele CCR5-modified hiPSCs from human lung fibroblasts and human primary cord blood mononuclear cells by site-specific insertion stem cell transcription factor genes flanked by LoxP sites into the endogenous CCR5 locus.     HLA-C-Dependent Prevention of Leukemia Relapse by Donor Activating KIR2DS1
    Researchers assessed clinical data, HLA genotyping results, and donor cell lines or genomic DNA for 1277 patients with acute myeloid leukemia who had received hematopoietic stem-cell transplants from unrelated donors matched for HLA-A, B, C, DR, and DQ or with a single mismatch.     Kruppel Like Factor 7 Overexpression Suppresses Hematopoietic Stem and Progenitor Cell Function
    Results suggest that Kruppel like factor 7 is not required for normal hematopoietic stem and progenitor function, but increased expression, as seen in a subset of lymphoid leukemia, inhibits myeloid cell proliferation and promotes early thymocyte survival. [Blood] Abstract

    CADASIL Mutations and shRNA Silencing of NOTCH3 Affect Actin Organization in Cultured Vascular Smooth Muscle Cells
    Researchers analyzed NOTCH3 expression and morphology of actin cytoskeleton in genetically genuine cultured human cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL) vascular smooth muscle cells (VSMCs) derived from different organs, and in control VSMCs with short hairpin RNA (shRNA)-silenced NOTCH3.     Vitamin D Improves the Angiogenic Properties of Endothelial Progenitor Cells
    Endothelial colony forming cells (ECFCs) were obtained and cultured from cord blood, and characterized by immunocytochemistry and flow cytometry. Proliferation, total length of tubule formation on Matrigel, expression of VEGF mRNA and proMMP-2 activity were assessed after treatment of ECFCs with vitamin D3.     SALL4 Is a Key Transcription Regulator in Normal Human Hematopoiesis
    Investigators found that SALL4 is a key regulator in normal human hematopoiesis and the mechanism of its function is at least in part through the HOXA9. Future study will determine whether modulating the SALL4/HOXA9 pathway can be used in cellular therapy such as cord blood expansion and/or myeloid engraftment. [Transfusion] Abstract
