Expansion of Human and Murine Hematopoietic Stem and Progenitor Cells Ex Vivo without Genetic Modification Using MYC and Bcl-2 Fusion Proteins
Scientists describe a novel approach to expand a cytokine-dependent hematopoietic stem and progenitor cell (HSPC) population ex vivo by culturing primary adult human or murine HSPCs with fusion proteins including the protein transduction domain of the HIV-1 transactivation protein and either MYC or Bcl-2. HSPCs obtained from either mouse bone marrow, human cord blood, human G-CSF mobilized peripheral blood, or human bone marrow were expanded. [PLoS One] Full Article | Press Release

PUBLICATIONS (Ranked by impact factor of the journal)
Transmissible Cytotoxicity of Multiple Myeloma Cells by Cord Blood-Derived NK Cells Is Mediated by Vesicle Trafficking
Researchers previously reported the encouraging observation that cord blood-derived natural killer cells (CB-NK), a new source of NK, showed anti-tumor activity in an in vivo murine model of multiple myeloma (MM) and confirmed a correlation between high levels of NKG2D expression by MM cells and increased efficacy of CB-NK in reducing tumor burden. They now characterize the mechanism of CB-NK-mediated cytotoxicity against MM cells. [Cell Death Differ] Abstract

Umbilical Cord Mesenchymal Stem Cells Inhibit the Differentiation of Circulating T Follicular Helper Cells in Patients with Primary Sjögren’s Syndrome through the Secretion of Indoleamine
    2,3-Dioxygenase

Scientists investigated the effect of umbilical cord mesenchymal stem cells (UC-MSCs) on circulating T follicular helper (cTfh) cells in primary Sjögren’s syndrome patients. In vitro, UC-MSCs suppressed the differentiation and proliferation of cTfh cells. [Rheumatology] Abstract

Treatment of Adult T-Cell Leukemia/Lymphoma with Cord Blood Transplantation: A Japanese Nationwide Retrospective Survey
To evaluate the role of cord blood transplantation (CBT) against adult T-cell leukemia/lymphoma (ATLL), investigators retrospectively analyzed data from 175 patients with ATLL who initially underwent single-unit CBT. [Biol Blood Marrow Transplant] Abstract

Human Bone Marrow, Umbilical Cord or Liver Mesenchymal Stromal Cells Fail to Improve Liver Function in a Model of CCl4-Induced Liver Damage in NOD/SCID/IL-2Rγ(Null) Mice
Researchers examined the effect of mesenchymal stromal cell (bone marrow, umbilical cord and liver MSCs) intravenous injection on liver regeneration in a model of hepatic damage in NOD/SCID/IL non-obese diabetic/severe combined immunodeficient/interleukin-2Rγ(null) mice. [Cytotherapy] Abstract

Myeloablative Unrelated Cord Blood Transplantation for Philadelphia Chromosome-Positive Acute Lymphoblastic Leukemia: Comparison with Other Graft Sources from Related and Unrelated Donors
Investigators retrospectively compared clinical outcomes after unrelated cord blood transplantation, unrelated bone marrow transplantation, and related bone marrow and peripheral blood stem cell transplantations following myeloablative conditioning in 40 patients with Philadelphia chromosome-positive acute lymphoblastic leukemia. [Ann Hematol] Abstract

Trisomy 21 Mid-Trimester Amniotic Fluid Induced Pluripotent Stem Cells Maintain Genetic Signatures during Reprogramming: Implications for Disease Modeling and Cryobanking
Researchers aimed to derive induced pluripotent stem cells from trisomy 21 human mid-trimester amniotic fluid stem cells and describe their hematopoietic and neurological characteristics. [Cell Reprogram] Abstract

miR-375 Induces Human Decidua Basalis-Derived Stromal Cells to Become Insulin-Producing Cells
miR-375 is a micro RNA that is expressed in the pancreas and involved in islet development. Human placental decidua basalis mesenchymal stem cells (PDB-MSCs) were cultured from full-term human placenta. The MSCs were chemically transfected with hsa-miR-375. [Cell Mol Biol Lett] Abstract

Mesenchymal Stem Cell-Based Angiotensin-Converting Enzyme 2 in Treatment of Acute Lung Injury Rat Induced by Bleomycin
After the isolation of human umbilical cord mesenchymal stem cells (HUMSCs) from cesarean fetus, scientists transfected the HUMSCs with angiotensin-converting enzyme 2 (ACE2) viral particles. They evaluated the therapeutic effect of HUMSCs harboring ACE2 on acute lung injury, which was induced by bleomycin in a rat model. [Exp Lung Res] Abstract

    INDUSTRY NEWS
Cord Blood Bank under FDA Lens for Operating without Licence
An ISO-certified cord blood bank in Sangli was found to be grossly negligent in labeling, testing and storing of stem cells. The Food and Drug Administration (FDA), which uncovered the violation last week, is now contemplating police action against the center and a crackdown on more such set-ups across state. [The Times of India] Editorial

AFL Approves Use of Regeneus’ Stem Cell Therapy, HiQCell for Injured Players
Regeneus Ltd. announced that the Australian Football League (AFL) has granted case-by-case approval for the use of its innovative stem cell therapy, HiQCell® as a treatment option for injured AFL players, typically including impact related osteoarthritis and tendonitis.
