Cryopreservation of Cord Blood CD34+ Cells before or after Thrombopoietin Expansion Differentially Affects Early Platelet Recovery in NOD SCID Mice
Investigators evaluated whether the order of cryopreservation and thrombopoietin expansion affected the expansion rate and numbers of clonogenic hematopoietic progenitor cells in vitro or platelet and longer-term hematopoietic repopulation in NOD SCID mice in vivo. [Transfusion] Abstract

Generation of Human Memory Stem T Cells upon Haploidentical T-Replete Hematopoietic Stem Cell Transplantation
Scientists found that donor-derived memory stem T cells (TSCM) cells are highly enriched early after hematopoietic stem cell transplantation. They showed at the antigen-specific and clonal level that TSCM lymphocytes can differentiate directly from naïve precursors infused within the graft and that the extent of TSCM generation might correlate with IL-7 serum levels. [Blood] Abstract

N-Butylidenephthalide Attenuates Alzheimer’s Disease-Like Cytopathy in Down Syndrome Induced Pluripotent Stem Cell-Derived Neurons
To recapitulate an Alzheimer’s disease cell model, Down syndrome induced pluripotent stem cells, reprogrammed from mesenchymal stem cells in amniotic fluid, were directed toward a neuronal lineage.

Plerixafor on Demand in Ten Healthy Family Donors as a Rescue Strategy to Achieve an Adequate Graft for Stem Cell Transplantation
Researchers report ten healthy hematopoietic stem cell donors treated with plerixafor because of insufficient response to granulocyte-colony-stimulating (G-CSF) alone or contraindication to G-CSF. [Transfusion] Abstract

MicroRNA-21 Promotes Osteogenic Differentiation of Mesenchymal Stem Cells by the PI3K/β-Catenin Pathway
MicroRNA-21 (miR-21) mimics and the antisense miR-21 were transfected to human umbilical cord mesenchymal stem cells (hUMSCs), and the capacity of miR-21 for the osteogenic differentiation of hUMSCs was evaluated by expression of osteogenic markers encoding alkaline phosphatase, runt-related gene-2 and osteocalcin, as well as by Alizarin red S staining. [J Orthop Res] Abstract

Co-Infusion of Mesenchymal Stromal Cells Has No Effect on Relapse and Infection in Leukemia Patients Undergoing Haploidentical Hematopietic Stem Cell Transplantation
Investigators observed the outcomes of mesenchymal stromal cells co-infusion during haploidentical hematopietic stem cell transplantation (haplo-HSCT) and compared the relapse and infection rates with patients receiving traditional haplo-HSCT. [Leuk Lymphoma] Abstract

Conditioned Medium from Umbilical Cord Mesenchymal Stem Cells Induces Migration and Angiogenesis
Scientists hypothesized that the paracrine effects of umbilical cord mesenchymal stem cells may enhance stem cell-based tissue repair and regeneration by promoting the specific homing of stem/progenitor cells and the overall ability to drive them to the damaged area. One Million Hemopoietic Stem-Cell Transplants: A Retrospective Observational Study
The authors aimed to document the activity worldwide from the beginning of transplantation and search for region adapted indications and associations between transplant rates and macroeconomics. [Lancet Haematol]

Allogeneic Hematopoietic Stem Cell Transplantation for Primary Myelofibrosis and Myelofibrosis Evolved from Other Myeloproliferative Neoplasms
Improvements in the field of allogeneic hematopoietic stem cell transplantation (allo-HSCT), the ability to improve patient’s performance status prior to transplant with JAK1/2 inhibitors and a more accurate disease risk stratification based on molecular mutations to select patients who can benefit from allo-HSCT should result in better transplant outcomes. UW to Test ‘Breakthrough’ Pediatric Leukemia Treatment
A promising method of immunotherapy to treat children with relapsed acute lymphocytic leukemia (ALL) is opening at the University of Wisconsin Carbone Cancer Center and American Family Children’s Hospital. The trial is open to relapsed/refractory pediatric ALL patients who have limited treatment options. [University of Wisconsin] Press Release
