Activated KRAS Cooperates with MLLAF4 to Promote Extramedullary Engraftment and Migration of Cord Blood CD34+ HSPC but Is Insufficient to Initiate Leukemia
Researchers explored whether KRAS activation cooperates with MLL-AF4 to initiate leukemia in cord blood-derived CD34+ hematopoietic stem/progenitor cells. Clonogenic and differentiation/proliferation assays demonstrated that KRAS activation does not cooperate with MA4 to immortalize CD34+ HSPCs. [Cancer Res] Abstract

Human Stem Cells Overexpressing miR-21 Promote Angiogenesis in Critical Limb Ischemia by Targeting CHIP to Enhance HIF-1α Activity
Umbilical cord blood-derived mesenchymal stem cells (UCBMSCs) were transduced with lentivirus-miR-21-Luciferase or lentivirus-LacZ-Luciferase. The results indicated that miR-21 induced UCBMSC proliferation, migration and angiogenesis in vitro. [Stem Cells] Abstract

Enhanced Renoprotective Effect of IGF-1 Modified Human Umbilical Cord-Derived Mesenchymal Stem Cells on Gentamicin-Induced Acute Kidney Injury
Scientists investigated whether insulin-like growth factor-1 (IGF-1)-modified umbilical cord-derived mesenchymal stem cells hold an enhanced protective effect on gentamicin-induced acute kidney injury in vivo. [Sci Rep] Full Article

The Generation of Human Innate Lymphoid Cells Is Influenced by the Source of Hematopoietic Stem Cells and by the Use of G-CSF
Scientists investigated the innate lymphoid cell differentiation potential of HSCs isolated from BM, mobilized peripheral blood, and umbilical cord blood. [Eur J Immunol] Abstract

Microcavity Arrays as an In Vitro Model System of the Bone Marrow Niche for Hematopoietic Stem Cells
Researchers established a three-dimensional (3D) co-culture system based on a custom-made chip, the 3D-KITChip, as an in vitro model system of the human hematopoietic stem cell niche. The microcavities of the 3D-KITChip were inoculated with human bone marrow mesenchymal stromal cells together with umbilical cord blood hematopoietic progenitor cells. [Cell Tissue Res] Abstract

BET Protein Inhibitor JQ1 Inhibits Growth and Modulates WNT Signaling in Mesenchymal Stem Cells
Scientists employed umbilical cord derived mesenchymal stem cells as a model system to evaluate the safety of JQ1. They showed that JQ1 inhibited cell growth and caused cell cycle arrest in G1 phase but did not induce apoptosis or senescence. [Stem Cell Res Ther] Full Article

Overexpression of CD61 Promotes hUC-MSC Differentiation into Male Germ-Like Cells
Researchers explored whether overexpression of CD61 would promote human umbilical cord mesenchymal stem cell (hUC-MSC) differentiation into primordial germ-like cells and male germ-like cells. [Cell Prolif] Abstract

The Effects of Indoxyl Sulfate on Human Umbilical Cord-Derived Mesenchymal Stem Cells In Vitro
Investigators aimed to determine the harmful effects of indoxyl sulfate on the phenotype and functional potential of human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) in vitro. They demonstrated that the immunophenotype of hUC-MSCs was not affected by indoxyl sulfate flow cytometry. [Cell Physiol Biochem] Full Article

Overexpression of Hypoxia-Inducible Factor-1 Alpha Improves Vasculogenesis-Related Functions of Endothelial Progenitor Cells
The authors overexpressed Hif-1α in peripheral blood endothelial progenitor cells (EPCs) and cord blood EPCs by using recombinant adenoviruses and investigated effects on stem cell- and vasculogenesis-related cell parameters. [Microvasc Res] Abstract

Modulation of microRNAs Expression in Hematopoietic Stem Cells Treated with Sodium Butyrate in Inducing Fetal Hemoglobin Expression
Investigators compared the changes in the microRNAs of differentiated erythroid cells between control and sodium butyrate treated groups. [Artif Cells Nanomed Biotechnol] Abstract
