TOP STORYThe authors studied the impact of HLA-DPB1 mismatch, along with the effect of other HLA-loci mismatches at the allele level. HLA six loci alleles were retrospectively typed in 1157 Japanese donors and patients with leukemia or myelodysplastic syndrome who underwent transplantation with a single unit of cord blood. PUBLICATIONS(Ranked by impact factor of the journal)Scientists investigated the role of nuclear factor erythroid-derived 2 (NF-E2) in normal human hematopoiesis. Knockdown of NF-E2 in the hematopoietic stem and progenitor cells not only reduced the formation of megakaryocytes but also drastically impaired hematopoietic stem cell activity, decreasing human engraftment in immunodeficient mice. [Stem Cell Reports] Full Article Researchers compared cord blood transplantation (CBT) outcomes in patients given single or double CBT following reduced-intensity conditioning (RIC) in a retrospective multicenter registry-based study. In univariate analysis, in comparison to patients transplanted with a single CB, double CB recipients had a similar incidence of neutrophil engraftment but a suggestion for a higher incidence of grade II–IV acute GVHD. [J Hematol Oncol] Full Article The authors designed a strategy to modify hepatocellular carcinoma (HCC) cells by surface expressing anti-CD3scfv within the tumor site strictly, which depended on the E1A-engineered human umbilical cord mesenchymal stem cells delivery system. Subsequently, membrane-bound anti-CD3scfv actived the lymphocytes which lysed HCC cells bypassing the expression of antigens or MHC restriction. [Int J Cancer] Abstract Investigators report that human induced human regulatory T cells generated in vitro from naïve cord blood cells preferentially recruit Disc large homolog 1 (Dlgh1) and exclude protein kinase C-θ from immunological synapses formed on supported lipid bilayers with laterally mobile ICAM-1 and anti-CD3 mAb. [Sci Rep] Full Article Mesenchymal Stem Cells Secretome-Induced Axonal Outgrowth Is Mediated by BDNF Scientists demonstrated that application of MSCs secretome to both rat cortical and hippocampal neurons induces an increase in axonal length. In addition, they showed that this growth effect is axonal intrinsic with no contribution from the cell body. [Sci Rep] Full Article Researchers illustrated that human umbilical cord-, bone marrow-, and adipose tissue-derived MSCs exhibited fibroblast-like morphology and expressed characteristic markers. Polycaprolactone nanofiber scaffold could support those MSC adhesion and proliferation. [Stem Cell Res Ther] Full Article Investigators aimed to determine whether human umbilical cord stem cell-conditioned medium (HUMSC-CM) improves the development of frozen-thawed in–vitro cultured ovarian tissues compared with a serum-free culture medium (SF-CM). A significant difference in the rate of morphologically normal primordial follicles in the HUMSC-CM group was observed compared to that in the serum-free culture medium group from days two to four. [Stem Cell Res Ther] Full Article Unrelated umbilical cord blood transplantation (UCBT) provides a readily available alternative donor source for patients lacking matched related donors. Scientists report the results of UCBT in 14 children treated between 1998 and 2016. [Biol Blood Marrow Transplant] Abstract Phase II Study of Intrabone Single Unit Cord Blood Transplantation for Hematological Malignancies In this Phase II study, 21 adult patients with hematological malignancy received intrabone transplantation of serological HLA-A, B, and DR ≥4/6 matched single cord blood with a median number of cryopreserved total nucleated cells of 2.7 × 107/kg following non-irradiated fludarabine-based reduced-intensity conditioning. [Cancer Sci] Full Article Scientists studied the reconstitution of natural killer (NK) cells and KIR-L mismatch in 54 patients who underwent a reduced intensity conditioning (RIC)-unrelated cord blood transplantation (UCBT) for acute myeloid leukemia in CR in a prospective Phase II clinical trial. After RIC-UCBT, NK cells displayed phenotypic features of both activation and immaturity. [Bone Marrow Transplant] Abstract | |

POLICY NEWSCanada’s Basic Science at Risk of Fading Away, Report Argues Canada’s scientific enterprise is at risk of sinking to junk bond status as a result of a funding crunch and misguided government and granting council policies, argues a report. In particular, the authors decry a steady shift away from basic science that has left a smaller portion of the nation’s growing scientific community focused on fundamental research. [ScienceInsider] Editorial Putin Tightens Control over Russian Academy of Sciences The Russian government has taken further steps to tighten its grip on the Russian Academy of Sciences (RAS) in Moscow. The State Duma—one of the two chambers of the Russian parliament—passed the first draft of a new law that would give president Vladimir Putin the final say in the elections for RAS’s presidency.
