    Hair Thinning by Stem Cell Loss
    To better understand the role hair follicle stem cells might play in aging-associated hair loss, scientists studied hair follicles in a mouse model of accelerated hair loss. An analysis of the animals’ skin at 18 months, a stage when their hair loss begins, showed that their hair follicles were miniaturized and reduced in number and thickness.     Epidermal β-Catenin Activation Remodels the Dermis via Paracrine Signaling to Distinct Fibroblast Lineages
    Sustained epidermal Wnt/β-catenin signaling expands the stem cell compartment and induces ectopic hair follicles. This is accompanied by extensive fibroblast proliferation and extracellular matrix remodeling in the underlying dermis.     Hierarchy of Cellular Decisions in Collective Behavior: Implications for Wound Healing
    To determine which individual cell behaviors represent the most promising targets to engineer re-epithelialization, investigators examined collective and individual responses of HaCaT keratinocytes seeded upon polyacrylamide gels of three stiffnesses and treated with a range of epidermal growth factor doses.     Comparative Transcriptomic Profiling of Hydrogen Peroxide Signaling Networks in Zebrafish and Human Keratinocytes: Implications toward Conservation, Migration and Wound Healing
    Comparative transcriptome analysis using RNA-seq data from larval zebrafish and previously reported microarray data from a human epidermal keratinocyte line showed that H2O2 activates conserved cell migration, adhesion, cytoprotective and anti-apoptotic programs in both zebrafish and human keratinocytes.     Nitric Oxide Chemical Donor Affects the Early Phases of In Vitro Wound Healing Process
    An artificial wound in a confluent monolayer of human keratinocyte HaCaT cells or mouse embryo fibroblast Swiss NIH 3T3 cells was used to analyze the effects of the nitric oxide chemical donor, S-nitroso-N-acetylpenicillamine. [J Cell Physiol] Abstract

    Self-Targeting of TNF-Releasing Cancer Cells in Preclinical Models of Primary and Metastatic Tumors
    The authors genetically engineered mouse mammary adenocarcinoma, melanoma, and Lewis lung carcinoma cells to produce and release murine tumor necrosis factor alpha (TNF).     Primary and Metastatic Cutaneous Melanomas Express ALK through Alternative Transcriptional Initiation
    Researchers determined whether anaplastic lymphoma kinase (ALK) was expressed in nonspitzoid primary and metastatic cutaneous melanomas. [Am J Surg Pathol] Abstract

    Phosphorylation of eIF4E Serine 209 Is Associated with Tumor Progression and Reduced Survival in Malignant Melanoma
    Researchers determined the association of eukaryotic translation initiation factor 4E (eIF4E) and phospho-eIF4E expression in melanocytic lesions with malignant conversion, metastatic potential and patient survival. [Br J Cancer] Abstract

    Pluripotency Markers Are Differentially Induced by MEK Inhibition in Thyroid and Melanoma BRAFV600E Cell Lines
    Cell line models of melanoma and thyroid carcinoma were treated with the MEK inhibitor PD0325901. Treated and naive samples were assayed for expression of key members of the MAPK pathway. [Cancer Biol Ther] Abstract

    Advances in Immunotherapy for Melanoma
    The combination of ipilimumab and nivolumab has yielded higher response rates, greater tumor shrinkage, and longer progression-free survival than either monotherapy alone. As other promising immunotherapies for melanoma proceed through clinical trials, future goals include defining the role of immune checkpoint inhibitors as adjuvant therapy, identifying optimal combination strategies, and developing reliable predictive biomarkers to guide treatment selection for individual patients.
