Melanoma Cells Block PEDF Production in Fibroblasts to Induce the Tumor-Promoting Phenotype of Cancer-Associated Fibroblasts
The authors demonstrated that normal dermal fibroblasts expressing high pigment epithelium-derived factor (PEDF) levels attenuated melanoma growth and angiogenesis in vivo, whereas PEDF-depleted fibroblasts exerted tumor-promoting effects. [Cancer Res] Abstract

Strong Components of Epigenetic Memory in Cultured Human Fibroblasts Related to Site of Origin and Donor Age
The authors previously identified relatively greater success culturing dura-derived fibroblasts than scalp-derived fibroblasts from postmortem tissue. They hypothesized that these differences in culture success were related to epigenetic differences between the cultured fibroblasts by sampling location, and therefore generated genome-wide DNA methylation and transcriptome data on 11 intrinsically matched pairs of dural and scalp fibroblasts from donors across the lifespan. Anti-Microbial Dendrimers against Multidrug-Resistant P. aeruginosa Enhance the Angiogenic Effect of Biological Burn-Wound Bandages
Anti-microbial polycationic dendrimers G3KL and G3RL, able to kill multi-drug resistant Pseudomonas aeruginosa, have been previously developed. The combination of these dendrimers with a class of biological bandages made of progenitor skin cells, which secrete growth factors, could positively impact wound-healing processes. Glycerolized Reticular Dermis as a New Human Acellular Dermal Matrix: An Exploratory Study
Scientists aimed to develop a new type of human acellular dermal matrix for clinical use, composed of glycerolized reticular dermis decellularized through incubation and tilting in Dulbecco’s Modified Eagle’s Medium. [PLoS One] Full Article

DNMT3B Modulates Melanoma Growth by Controlling Levels of mTORC2 Component RICTOR
Scientists report that DNMT3B plays a pro-tumorigenic role in human melanoma and that DNMT3B loss dramatically suppresses melanoma formation in the Braf/Pten mouse melanoma model. [Cell Rep]

Melatonin Protects Skin Keratinocyte from Hydrogen Peroxide-Mediated Cell Death via the SIRT1 Pathway
The authors investigated the protection effect of melatonin on H2O2-induced cell damage and the mechanisms of melatonin in human keratinocytes. [Oncotarget] Full Article

Ultraviolet Radiation-Induced Tumor Necrosis Factor Alpha, Which Is Linked to the Development of Cutaneous SCC, Modulates Differential Epidermal MicroRNAs Expression
To find clues about the mechanisms by which tumor necrosis factor-alpha may promote ultraviolet radiation-induced development of squamous cell carcinoma (SCC), researchers investigated changes in the expression profiling of microRNAs, which affects translation and stability of mRNAs. [Oncotarget] Full Article

Transketolase-Like 1 Ectopic Expression Is Associated with DNA Hypomethylation and Induces the Warburg Effect in Melanoma Cells
Investigators evaluated the role of transketolase-like 1 as a mediator of disease progression in melanoma associated with a defective methylation phenotype. [BMC Cancer] Full Article

Solar Simulated Ultraviolet Radiation Induces Global Histone Hypoacetylation in Human Keratinocytes
Investigators report global changes in histone post-translational modifications induced by solar simulated ultraviolet radiation (ssUVR). A decrease in lysine acetylation of histones H3 and H4, particularly at positions of H3 lysine 9, lysine 56, H4 lysine 5, and lysine 16, was found in human keratinocytes exposed to ssUVR. Histone Variants and Melanoma: Facts and Hypotheses
The authors focus on the role of histone variants, key epigenetic players in melanoma initiation and progression. Specifically, incorporation of histone variants enables additional layers of chromatin structure and they describe how alterations in this epigenetic behavior impact melanoma.
