    Scientists Achieve Breakthrough in Diagnosis and Therapy of Melanoma Skin Cancer
    Scientists revealed a remarkable link between malignant melanoma and a non-coding RNA gene called SAMMSON. The SAMMSON gene is specifically expressed in human malignant melanoma and, strikingly, the growth of aggressive skin cancer is highly dependent on this gene.     Multicolor Cell Barcoding Technology for Long-Term Surveillance of Epithelial Regeneration in Zebrafish
    Researchers employed skinbow-based imaging in conjunction with a live reporter of epithelial stem cell cycle activity and as an instrument to evaluate the effects of reactive oxygen species on superficial epithelial cell behavior during epithelial regeneration. They introduced a platform for large-scale, quantitative in vivo imaging of regenerating skin and revealed unanticipated collective dynamism in epithelial cell size, mobility, and interactions.     Perlecan Expression Influences the Keratin 15-Positive Cell Population Fate in the Epidermis of Aging Skin
    The authors revealed reduced perlecan transcript levels in aged keratinocytes. The production of in vitro skin models revealed that aged keratinocytes formed a thin and poorly organized epidermis. [Aging (Albany NY)] Full Article

    5α-Dihydrotestosterone Enhances Wound Healing in Diabetic Rats
    Researchers analyzed the effects of 5α-dihydrotestosterone ointment in cutaneous wound healing by secondary intention in diabetic Wistar rats. [Life Sci] Abstract

    Enhanced Cancer Immunotherapy by Microneedle Patch-Assisted Delivery of Anti-PD1 Antibody
    Investigators report an innovative self-degradable microneedle (MN) patch for the sustained delivery of anti-PD-1 (aPD1) in a physiologically controllable manner. They found that a single administration of the MN patch induces robust immune responses in a B16F10 mouse melanoma model compared to MN without degradation trigger or intratumoral injection of free aPD1 with the same dose.     NFAT1 Directly Regulates IL-8 and MMP-3 to Promote Melanoma Tumor Growth and Metastasis
    Researchers report a strong correlation between nuclear factor of activated T cell (NFAT1) expression and metastatic potential in melanoma cell lines and tumor specimens. To elucidate the mechanisms underlying NFAT1 overexpression during melanoma progression, they conducted a microarray on a highly metastatic melanoma cell line in which NFAT1 expression was stably silenced.     Long-Lasting Complete Responses in Patients with Metastatic Melanoma after Adoptive Cell Therapy with Tumor-Infiltrating Lymphocytes and an Attenuated IL-2 Regimen
    Interleukin-adoptive cell transfer therapy with a reduced interleukin-2 (IL-2) decrescendo regimen resulted in long-lasting complete responses in patients with treatment-refractory melanoma. [Clin Cancer Res] Abstract

    Suppression of Type I Interferon Signaling Overcomes Oncogene-Induced Senescence and Mediates Melanoma Development and Progression
    Researchers report a key role of type I interferons (IFNs) in oncogene-induced senescence. IFN signaling-deficient melanocytes expressing activated Braf did not exhibit senescence and develop aggressive melanomas. [Cell Rep] Full Article | Press Release

    Inactivation of RASA1 Promotes Melanoma Tumorigenesis via R-Ras Activation
    Scientists identified two novel, clustered somatic missense mutations in RASA1. They have shown that wild type RASA1, but not identified mutants, suppressed soft agar colony formation and tumor growth of BRAF mutated melanoma cell lines via its RasGAP activity toward R-Ras isoform.     HOPX: The Unusual Homeodomain-Containing Protein
    In human skin, homeodomain-only protein homeobox (HOPX) controls epidermal formation through the regulation of late differentiation markers, and HOPX expression correlates with the level of differentiation in cutaneous pathologies. In mouse skin, Hopx was additionally identified as a lineage tracing marker of quiescent hair follicle stem cells.
