Tumor volumes were measured and samples from the NRAS-mutant melanomas were collected for immune-cell analysis, single-cell RNA sequencing, and reverse phase protein analysis.

    MTH1 Inhibitors for the Treatment of Psoriasis

    Using the Imiquimod psoriasis mouse model, researchers found that MutT homolog 1 (MTH1) inhibition diminished psoriatic histological characteristics and normalized the levels of neutrophils and T cells in skin and skin-draining lymph nodes. The inhibition abolished the expression of Th17-associated cytokines in the skin, which was in line with decreased levels of IL-17-producing γδ T cells in lymph nodes.

    Scientists engineered an Olmsted syndrome mouse model by introducing the point mutation G568V to the corresponding Trpv3 locus in mice. These mice developed fully penetrant hair loss.

    Out Alpha-Synuclein in Melanoma Cells Dysregulates Cellular Iron Metabolism and Suppresses Tumor Growth

    To probe the function of alpha-synuclein (α-syn) in melanoma, the authors used CRISPR/Cas9 to knockout SNCA, the gene that codes for protein α-syn, in SK-Mel-28 melanoma cells. The SNCA-knockout clones in culture exhibited a decrease in the transferrin receptor 1, an increase in ferritin, an increase of reactive oxygen species and proliferated slower than control cells.

    Granzyme B Mediates Impaired Healing of Pressure Injuries in Aged Skin

    A causative role for Granzyme B (GzmB) was assessed in GzmB knockout and wild-type mice using a murine model of pressure injuries. An apolipoprotein E knockout model of aging and vascular dysfunction was also utilized to assess GzmB in a relevant age-related model better resembling tissue perfusion in the elderly.
