TOP STORYFunctional Importance of a Proteoglycan Co-Receptor in Pathologic Lymphangiogenesis Lymphatic endothelial deficiency in the major heparan sulfate biosynthetic enzyme N-deacetylase/N-sulfotransferase-1 was associated with reduced lymphangiogenesis in pathologic models, including spontaneous neoplasia. PUBLICATIONS(Ranked by impact factor of the journal)Von Willebrand Factor Regulates Complement on Endothelial Cells The authors investigated the possible functional link of von Willebrand factor (VWF)-multimers and the complement system on endothelial cells. Blood outgrowth endothelial cells were obtained from three healthy individuals and two patients with Type 3 von Willebrand disease lacking VWF. [Kidney Int] Abstract Scientists investigated the changes in hemogenic potential in endothelial progenitors at the early stages of embryonic development. Using an ETV2::GFP reporter mouse to isolate emerging endothelial progenitors, they observed a dramatic decrease in hemogenic potential between embryonic day (E)7.5 and E8.5. [Cell Rep] Full Article | Graphical Abstract In vitro, the involvement of the PI3K/Akt/eNOS pathway in the effect of U50,488H was studied using cultured endothelial cells subjected to artificial hyperlipidemia. [Sci Rep] Full Article This study investigated the therapeutic effects and mechanisms of Niaspan treatment of diabetic retinopathy in diabetes (DM) rats. DM rats exhibited significantly decreased microRNA-126 and tight junction Claudin-5/Occludin/ZO-1 genes expression, and increased blood retinal-barrier breakdown, retinal apoptosis and VEGF/VEGFR, as well as VCAM-1/CD45 expressions in the retina compared to normal control group. [Sci Rep] Full Article The authors hypothesized that a high-fat (HF) diet generates a dysfunctional vascular niche through an increased expression of endothelial Forkhead box O (FoxO). FoxO1 protein increased in the skeletal muscle capillaries from HF compared to normal chow-fed mice. FoxO1 protein was significantly elevated in cultured endothelial cells exposed to the saturated fatty acid palmitate or the proinflammatory cytokine TNF-α. [FASEB J] Abstract Investigators describe a model system utilizing oxidative lipidomics/mass spectrometry and bioassays on cultured bovine pulmonary artery endothelial cells to assess the impact of oxidized cardiolipin that they showed, in vivo, can be released to the extracellular space and may be hydrolyzed by lipoprotein-associated phospholipase A2. [Am J Physiol Lung Cell Mol Physiol] Abstract Researchers evaluated the interaction between tumor-associated macrophages and lymphatic endothelial cells (LECs) derived from lymph nodes of human gastric cancer. LECs were directly or indirectly co-cultured with macrophages from healthy human blood, with or without the supernatant of the gastric cancer cell line, OCUM-12. [Cancer Sci] Abstract The eicosanoid precursor arachidonic acid stimulated vasculogenesis of embryonic stem cells by increasing the number of fetal liver kinase-1+ vascular progenitor cells as well as vascular structures positive for platelet endothelial cell adhesion protein-1 and vascular endothelial cadherin. REVIEWSThe authors summarize the current knowledge of EphrinB2/EphB4 in angiogenesis and their interaction with other angiogenic pathways in ischemic cardiovascular disease. [Angiogenesis] Abstract The authors summarize the main roles and pathways controlled by ERG in the vascular endothelium, its transcriptional targets and its functional partners and the emerging evidence on the pathways regulating ERG’s activity and expression.
