TOP STORYSerine Synthesis via PHGDH Is Essential for Heme Production in Endothelial Cells Scientists reported that mouse neonates with endothelial cell (EC)-specific phosphoglycerate dehydrogenase (PHGDH) deficiency suffered lethal vascular defects within days of gene inactivation, due to reduced EC proliferation and survival. PUBLICATIONS(Ranked by impact factor of the journal)Using a combination of genetic and molecular approaches, researchers identified the transcription factor hematopoietically expressed homeobox (HHEX) as an upstream regulator of VEGFC, FLT4, and PROX1 during angiogenic sprouting and lymphatic formation in vertebrates. Studies of mammalian HHEX using tissue-specific genetic deletions in mouse and knockdowns in cultured human endothelial cells revealed its highly conserved function during vascular and lymphatic development. [Nat Commun] Full Article In HUVECs, polydatin inhibited VEGF-stimulated cell proliferation, cell migration, and tube formation. Moreover, polydatin showed suppressive effects on the subintestinal vessel formation in zebrafish embryos. [FASEB J] Abstract Endothelial progenitor cell adhesion, migration and in vitro tube formation were promoted by angiotensin II, and the effects were reversed by RhoA/Rho-associated kinases signaling pathway inhibitors including C3 exoenzyme, GGTI-286 and Y-27632. [Cell Physiol Biochem] Full Article Lung endothelial cells isolated from endothelium‐specific kallistatin knockout mice displayed marked reduction in mouse kallistatin levels. Kallistatin deficiency in mouse endothelial cells exacerbated senescence, oxidative stress and inflammation compared to wild‐type mouse endothelial cells, and H2O2 treatment further magnified these effects. [J Cell Mol Med] Abstract | Full Article Scientists investigated the importance of Ang-2 in nucleoside diphosphate kinase B (NDPK-B) deficient retinas and characterized the mechanisms of angiopoetin 2 upregulation upon NDPK-B depletion in cultured endothelial cells. [Sci Rep] Full Article Inhibition of Angiogenesis in Endothelial Cells by Human Lysyl Oxidase Propeptide The propeptide domain of LOX (LOX-PP) has been shown to inhibit tumorigenesis in various cancers. Investigators explored the role of both overexpressed and recombinant LOX-PP in naïve HUVECs with the addition of VEGF. [Sci Rep] Full Article miR-126-3p-overexpressing endothelial progenitor cells (MO-EPCs) or non-modified EPCs (NM-EPCs) were transplanted into nude rats with ischemic cardiomyopathy induced by coronary artery ligation. Eight weeks after MO-EPC transplantation, left ventricular function improved significantly with clearly decreased infarction size, increased anterior wall thickness, and inhibition of inflammation compared with the results for NM-EPC transplantation. [Circ J] Full Article Researchers investigated the role and molecular mechanisms of mammalian target of rapamycin complex 2 (mTORC2) in endothelial senescence. A replicative senescence model and H2O2-induced premature senescence model were established in primary cultured HUVECs. [Acta Pharmacol Sin] Abstract Mini-Sheet Injection for Cultured Corneal Endothelial Transplantation The authors reported a novel mini-sheet injection for the cultured corneal endothelial cell transplantation and compared it with the effects of single cell injection in a rabbit model. POLICY NEWSScience Journals End Open-Access Trial with Gates Foundation The publisher of Science ended a pilot partnership that allowed open-access (OA) publishing for researchers funded by the Bill & Melinda Gates Foundation. The trial was an effort to accommodate a policy clash between the Gates Foundation, which has enforced strict OA demands since 2017, and publishers that run subscription journals which don’t comply with those terms. [Nature News] Editorial Researchers Celebrate Rejection of Controversial Copyright Policy Early-career researchers are pleased at European leaders’ rejection of a copyright policy that would have introduced restrictions to text-and-data mining of scientific papers.
