TOP STORYNanomaterials-induced endothelial leakiness might favor intravasation of surviving cancer cells into the surrounding vasculature and subsequently extravasation, accelerating metastasis. Investigators showed that nanoparticles induced endothelial leakiness through disruption of the VE-cadherin-VE-cadherin homophilic interactions at the adherens junction. PUBLICATIONS(Ranked by impact factor of the journal)TAK1 Regulates Endothelial Cell Necroptosis and Tumor Metastasis Scientists identified endothelial TGF-β-activated kinase 1 (TAK1) as a critical regulator of endothelial necroptosis and metastasis. Human and murine endothelial cells lacking TAK1 exhibited higher levels of necroptosis both in vitro and in vivo, and mice with endothelial cell-specific loss of TAK1 were more prone to form metastases. [Cell Death Differ] Abstract Investigators demonstrated that aged cardiac microvascular endothelial cells utilized BDNF-TrkB-T1 signaling to recruit Willin as a downstream effector to activate the Hippo pathway, which then promotes migration. [Aging Cell] Full Article Mechanistic studies using transcriptomic profiling, bioinformatics, 3′-UTR reporter and miRNA ribonucleoprotein complex -immunoprecipitation assays, and small interfering RNA dependency studies revealed that miR-135a-3p inhibited the p38 signaling pathway in endothelial cells by targeting huntingtin-interacting protein 1. [FASEB J] Abstract Nerve Growth Factor Regulates Endothelial Cell Survival and Pathological Retinal Angiogenesis In vitro, nerve growth factor (NGF) decreased the intrinsic apoptosis in hypoxia-treated human retinal microvascular endothelial cells and preserved the mitochondrial membrane potential. The anti-apoptotic effect of NGF was associated with increased BCL2 and reduced BAX, as well as with enhanced ERK and AKT phosphorylation, and was abolished by inhibition of the AKT pathway. [J Cell Mol Med] Full Article Stem Cell Exosomes Inhibit Angiogenesis and Tumor Growth of Oral Squamous Cell Carcinoma Researchers showed that different types of endothelial cells presented specific kinetics for exosome internalization. Exosome-treatment of endothelial cells increased cytotoxicity and reduced VEGF secretion and angiogenesis in a dose-dependent manner. [Sci Rep] Full Article Epidermal Growth Factor Receptor Promotes Cerebral and Retinal Invasion by Toxoplasma gondii Parasite foci in brain endothelial cells were reduced by dominant negative (DN) EGFR. DN EGFR in these cells led to recruitment of the autophagy protein LC3 around T. gondii and spontaneous parasite killing dependent on the autophagy protein ULK1 and lysosomal enzymes. [Sci Rep] Full Article A recombinant protein expressing only the extracellular portion of delta-like ligand 4 (Dll4) induced Notch signaling in endothelial cells, resulting in increased expression of VE-cadherin, but not the tight junctional protein ZO1, at intercellular junctions. [Am J Physiol Heart Circ Physiol] Abstract Investigators showed that a high methionine diet induced hyperhomocysteinemia, impaired both adhesion and angiogenesis properties of bone marrow endothelial progenitor cells, accompanied by a higher methylation level of the cystathionine-β-synthase (CBS) promoter compared to the control. [Biochem Biophys Res Commun] Abstract Blocking C/EBP β Protects Vascular Endothelial Cells from Injury Induced by Intermittent Hypoxia Blocking CCAAT/enhancer binding protein beta (C/EBP β) impaired intermittent hypoxia-induced apoptosis but did not affect VEGF expression, proliferation, or in vitro tube formation. C/EBP β was shown to mediate increased endothelin-1 (ET-1) transcription by hypoxia inducible factor-1 alpha through the transcription factor binding site, 5′-GTTGCCTGTTG-3′, in the ET-1 promoter. INDUSTRY NEWSSTEMCELL Technologies has released mTeSR™ Plus, an enhanced version of mTeSR™1, the most widely published feeder-free human PSC maintenance medium. The new medium has undergone successful beta testing with several members of the Stem Cell COREdinates group and has completed rigorous in-house validation testing. [STEMCELL Technologies Inc.] Press Release Efforts will now be exclusively directed to file integrated regulatory submissions in 2020 with the European Medicines Agency and FDA for full marketing approval, after the planned release of topline data from the Phase III ADVOCATE clinical trial anticipated already in the fourth quarter of 2019. [ChemoCentryx, Inc.] Press Release EMA Staff Lower Flags at London Office as Amsterdam Move Nears Staff at the European Medicines Agency (EMA) have lowered the flags outside their London offices as they prepare to move to Amsterdam. The flag lowering took place as the EMA “symbolically said goodbye” to the city it has called home since its creation in 1995. [Questex LLC.] Press Release | |

POLICY NEWSKosovo Academics Brace for Controversial Ethnic Land Swaps A proposed land swap between Serbia and Kosovo, which is designed to help end a decades-long ethnic conflict in the region, could threaten rare attempts at multi-ethnic research collaboration and education. The exchange aims to settle ownership of the remaining disputed areas between the two hostile neighbors, but it could also result in academic institutions finding themselves in a different, essentially enemy, country. [Nature News] Editorial US Science Agencies Reopen after Shutdown On January 25, lawmakers ended the longest partial government shutdown in US history and agreed to open federal agencies for three weeks as they negotiate President Donald Trump’s demand for a wall along the southern border.
