TOP STORYA Novel L1CAM Isoform with Angiogenic Activity Generated by NOVA2-Mediated Alternative Splicing Scientists report that endothelial cells (ECs) expressed a novel isoform of the cell-surface adhesion molecule L1CAM, termed L1-ΔTM. The splicing factor NOVA2, which bound directly to L1CAM pre-mRNA, was necessary and sufficient for the skipping of L1CAM transmembrane domain in ECs, leading to the release of soluble L1-ΔTM. PUBLICATIONS(Ranked by impact factor of the journal)Antibody ligation of human leukocyte antigen (HLA) class II molecules in interferon-γ-treated endothelial cells caused necrotic cell death without complement via a pathway that was independent of apoptosis and necroptosis. [Cell Death Dis] Full Article Engagement of lymphotoxin-β receptor (LTβR) triggered canonical and non-canonical NF-κB signaling, promoted expression of inflammatory mediators and adhesion molecules, and increased immune cell adhesion to endothelial cells (ECs). Sustained LTβR-induced inflammatory activation of ECs was NF-κB-inducing kinase (NIK)-dependent, but independent of p100, indicating the non-canonical arm of NF-κB was not involved. [J Cell Sci] Abstract Functional Characterization of iPSC-Derived Arterial- and Venous-Like Endothelial Cells Functionally, both arterial and venous-like induced pluripotent stem cell (iPSC)-derived endothelial cells (ECs) responded to vasoactive agonists such as thrombin and prostaglandin E2, similar to somatic ECs; however, arterial-like iPSC-derived ECs produced higher nitric oxide and elongation to shear stress than venous-like iPSC-derived ECs [Sci Rep] Full Article Researchers investigated whether a single cell label-free multi parameter optical interrogation system could detect endothelial cell and endothelial progenitor cell activation in vitro and ex vivo, respectively. [Sci Rep] Full Article The authors showed that PGE2 stimulated cAMP production and activated protein kinase A (PKA), whereas pretreatment with the adenyl cyclase inhibitor or PKA inhibitors, H89/PKAI, completely abrogated PGE2-induced migration. [J Leukoc Biol] Abstract Cinobufagin Suppresses Colorectal Cancer Angiogenesis by Disrupting Endothelial mTOR/HIF-1α Axis Endothelial apoptosis was observed by inducing reactive oxygen species accumulation and mitochondrial dysfunction which could be neutralized by N-acetyl-L-cysteine. The expression of hypoxia-inducible factor 1α (HIF-1α) was reduced and phosphorylation of mechanistic target of rapamycin (mTOR) at Ser2481 and Akt at Ser473 was downregulated in HUVECs. [Cancer Sci] Abstract The expression of miR-183, CD34, endothelial nitric oxide synthase, BTG1 and the PI3K/Akt/VEGF signaling pathway-related genes as well as reactive oxygen species level was determined, and the relationship between miR-183 and BTG1 was also verified. [Am J Physiol Endocrinol Metab] Abstract Investigators demonstrated that plasma collected from women who subsequently developed preeclampsia caused increased heme oxygenase-1 (HO-1) production and decreased levels of nitric oxide (NO) markers in endothelial cells. Conversely, no changes in HO-1 or NO markers were found when HUVECs were treated with plasma from women who remained healthy throughout pregnancy. [Hypertens Res] Abstract Overexpression of Axl Reverses Endothelial Cells Dysfunction in High Glucose and Hypoxia Axl overexpression significantly reversed endothelial cell dysfunction in high glucose/hypoxia. Furthermore, Axl overexpression in EAhy926 cells increased hypoxia-inducible factor 1-α protein synthesis through PI3K/Akt/mTOR/p70S6K signal pathway but not Mek/Erk in high glucose/hypoxia condition. [J Cell Biochem] Abstract Telbivudine Reduces Parvovirus B19-Induced Apoptosis in Circulating Angiogenic Cells Scientists tested the hypothesis of whether telbivudine influenced human parvovirus B19 (B19V)-induced apoptosis of circulating angiogenic cells (CACs). Pretreatment of two CAC-lines, early outgrowth endothelial progenitor cells and endothelial colony-forming cells with telbivudine before in vitro infection with B19V significantly reduced active caspase-3 protein expression.
