Comparative Transcriptomic Analysis Identifies a Range of Immunologically Related Functional Elaborations of Lymph Node Associated Lymphatic and Blood Endothelial Cells

Researchers used RNA-Seq to compare the transcriptomic profiles of freshly isolated murine lymphatic endothelial cells (LECs) and blood ECs (BECs) from lymph nodes (LNs) with one another and with freshly isolated LECs from the periphery. They showed that LN-LECs, LN-BECs, and diaphragm LECs were transcriptionally distinct from one another, demonstrating both lineage and tissue-specific functional specializations. Adiponectin Inhibits High Glucose-Induced Angiogenesis via Inhibiting Autophagy in RF/6A Cells

Treatment with adiponectin or 3-methyladenine (3-MA) increased cell viability and inhibited cell migration and tube formation. In the high-glucose group, the protein expression of Bax and apoptosis rate of cells increased and the expression of Bcl-2 decreased, whereas treatment with adiponectin or 3-MA reversed these results. Plasminogen activator inhibitor 1 (PAI-1)-siRNA enhanced the luminal formation ability of endothelial progenitor cells (EPCs) and significantly promoted EPC homing. In response to PAI-1 gene silencing, tissues from inferior vena cava displayed reduced mRNA and protein expression of PAI-1, increased VEGF expression as well as promoted lumen-like structures.
