miR-181a Delivered by Hypoxic Papillary Thyroid Cancer-Secreted Exosomes Inhibits DACT2 by Down-Regulating MLL3, Leading to YAP-VEGF Pathway-Mediated Angiogenesis

HUVECs were transfected with miR-181a inhibitor/mimic or siRNA-MLL3, or treated with exosomes from hypoxic papillary thyroid cancer cells. Hypoxic exosomal miR-181a delivery promoted proliferation and capillary-like network formation in HUVECs.

An In Vitro Vascularized Micro-Tumor Model of Human Colorectal Cancer Recapitulates In Vivo Responses to Standard-of-Care Therapy

Researchers present a vascularized micro-tumor that is a novel three-dimensional model system that recapitulated the complex human tumor microenvironment, including perfused vasculature, within a transparent microfluidic device, allowing real-time study of drug responses and tumor-stromal interactions.

Single-Cell Transcriptomic Profiling and Characterization of Endothelial Progenitor Cells: New Approach for Finding Novel Markers

Scientists assembled five groups of chosen endothelial progenitor cells (EPCs)-related genes/factors using PubMed literature and Gene Ontology databases. This shortened database of EPC factors was fed into publically published transcriptome matrix to compare their expression between endothelial colony-forming cells, HUVECs, and two adult endothelial cell types from the skin and adipose tissue.

CREB3L4 Promotes Angiogenesis and Tumor Progression in Gastric Cancer through Regulating VEGFA Expression

The upregulation of cyclic AMP responsive element-binding protein 3-like 4 (CREB3L4) in gastic cancer cells increased cell viability, promoted cell proliferation, reduced apoptosis, enhanced cell migration and invasion, and induced the formation of tubule-like endothelial structures, whereas CREB3L4 knockdown impeded tumor cell growth, attenuated cell motility, and prevented human umbilical vein endothelial cells from forming tubule-like structures.

Human Mesenchymal Stem Cells Promote Ischemic Repairment and Angiogenesis of Diabetic Foot through Exosome miRNA-21-5p

Exosomes were isolated from MSCs using ultracentrifugation, and further characterized by the nanoparticle tracking analyzer and flow cytometry. RNA sequencing, Western Blot, in vitro cell proliferation, in vivo pro-angiogenesis, as well as ischemic repairment of diabetic foot through rat model were performed to evaluate exosome physiological functions.

FGF21 Promotes Ischemic Angiogenesis and Endothelial Progenitor Cells Function under Diabetic Conditions in an AMPK/NAD+-Dependent Manner

The in vivo results showed that FGF21 efficiently improved blood perfusion and ischaemic angiogenesis in both type 1 and type 2 diabetic mice, and these effects were accompanied by enhanced endothelial progenitor cells mobilization and infiltration into ischemic muscle tissues and increases in plasma stromal cell–derived factor‐1 concentration.

The Cancer Glycocalyx Mediates Intravascular Adhesion and Extravasation during Metastatic Dissemination

Using an in vitro model of the human microvasculature, researchers assessed the importance of the tumor and vascular glycocalyces during tumor cell extravasation.

3D Bioprinting of Tissue-Specific Osteoblasts and Endothelial Cells to Model the Human Jawbone

Production of human jawbone models was realized via projection-based stereolithography. Constructs were bioprinted containing primary jawbone-derived osteoblasts and vasculature-like channel structures optionally harboring primary endothelial cells.

Tetraspanin18 Regulates Angiogenesis through VEGFR2 and Notch Pathways

The authors determined that Tspan18, a member of the Tetraspanin family, was expressed in developing vessels but not in mature vasculature in zebrafish and mouse wound healing. Reduction at Tspan18 level resulted in aberrant vascular patterning, impaired vessel stability and defective arterial–venous specification.

Strategies for Delivering Therapeutics across the Blood–Brain Barrier

Investigators review recent developments and challenges related to selected blood-brain barrier-crossing strategies- with a focus on non-invasive approaches such as receptor-mediated transcytosis and the use of neurotropic viruses, nanoparticles and exosomes- and analyze their potential in the treatment of central nervous system disorders.
