iPSC–Endothelial Cell Phenotypic Drug Screening and In Silico Analyses Identify Tyrphostin-AG1296 for Pulmonary Arterial Hypertension

Induced pluripotent stem cell-derived endothelial cells generated from six patients with pulmonary arterial hypertension were exposed to 4500 compounds and assayed for improved cell survival after serum withdrawal using a chemiluminescent caspase assay.

Sox9 and Rbpj Differentially Regulate Endothelial to Mesenchymal Transition and Wound Scarring in Murine Endovascular Progenitors

Skin wound analysis from Sox9 conditional knock-out mice demonstrated a significant reduction in pathological endothelial to mesenchymal transition (EndMT) resulting in reduced scar area. The converse was observed with Rbpj conditionally knocked-out from the murine vasculature or inhibition of Notch signaling in human endothelial colony forming cells, resulting in enhanced Sox9 and EndMT related gene

Notch Regulates Vascular Collagen IV Basement Membrane through Modulation of Lysyl Hydroxylase 3 Trafficking

Researchers identified trafficking pathways that regulated collagen type IV (Col IV) deposition during angiogenic blood vessel development. They identified the GTPase Rab10 as a major regulator of Col IV vesicular trafficking during vascular development using both in vitro imaging and biochemistry as well as in vivo models.

Role of Epithelial-Endothelial Cell Interaction in the Pathogenesis of SARS-CoV-2 Infection

Scientists measured the susceptibility to infection, immune response, and expression of adhesion molecules, in human pulmonary microvascular endothelial cells exposed to conditioned medium from infected epithelial cells.

    Abstract

Supramolecular Self-Assembled Nanofibers Efficiently Activate the Precursor of Hepatocyte Growth Factor for Angiogenesis in Myocardial Infarction Therapy

Investigators designed supramolecular nanofibers self-assembled by compound 1, which could strongly activate the hepatocyte growth factor precursor and initiate hepatocyte growth factor–Met signaling.

    Abstract

Mineralocorticoid and Estrogen Receptors in Endothelial Cells Coordinately Regulate Microvascular Function in Obese Female Mice

Estrogen receptor α (ERα) can oppose mineralocorticoid receptor (MR) function, therefore, scientists hypothesized that ERα mediated the benefits of endothelial MR deficiency.

A Novel Strategy for Isolation of Mice Bone Marrow Endothelial Cells (BMECs)

Researchers outlined a simple new technique of isolating mouse BMEC primary culture and a suitable method to evaluate the function and dysregulation of BMEC in in vitro studies using mouse models.

Baicalein Alleviates Osteoarthritis by Protecting Subchondral Bone, Inhibiting Angiogenesis and Synovial Proliferation

The authors found that intra‐articular injection of baicalein ameliorated subchondral bone remodeling. Baicalein impaired angiogenesis of endothelial cells and inhibited proliferation of synovial cells.

Neutrophil Oxidative Stress Mediates Obesity-Associated Vascular Dysfunction and Metastatic Transmigration

Scientists showed that obesity led to neutrophil-dependent impairment of vascular integrity through loss of endothelial adhesions, enabling cancer cell extravasation into the lung.

Fibroblast CEBPD/SDF4 Axis in Response to Chemotherapy-Induced Angiogenesis through CXCR4

Investigators found that cisplatin and 5-fluorouracil could induce fibroblast differentiation toward myofibroblasts via CCAAT/enhancer-binding protein delta and consequently promoted proliferation, migration, and in vitro tube formation of vascular endothelial cells and angiogenesis in vivo.

Interplay between Inflammation and Thrombosis in Cardiovascular Pathology

Scientists discuss how the synergy between inflammation and thrombosis drives thrombotic diseases. They focus on the huge potential of anti-inflammatory strategies to target cardiovascular pathologies.

High Endothelial Venules (HEVs) in Immunity, Inflammation and Cancer

The authors highlight the phenotype and function of high endothelial venules (HEVs) in homeostatic, inflamed and tumor-draining lymph nodes, and those of HEV-like blood vessels in chronic inflammatory diseases.
