    TOP STORY

Multispectral Fingerprinting Resolves Dynamics of Nanomaterial Trafficking in Primary Endothelial Cells

Scientists developed and applied an approach where single-walled carbon nanotubes played a dual role-that of an engineered nanomaterials undergoing intracellular processing, in addition to functioning as the signal transduction element reporting these events in individual cells with single organelle resolution.

Continuous Monitoring Reveals Protective Effects of N-Acetylcysteine Amide on an Isogenic Microphysiological Model of the Neurovascular Unit

Investigators reported the first model of the blood-brain barrier featuring both isogenic human induced pluripotent stem cell-derived cells and continuous barrier integrity monitoring with under 2 min temporal resolution.

LncRNA VEAL2 Regulates PRKCB2 to Modulate Endothelial Permeability in Diabetic Retinopathy

Using PRKCB2 as bait, scientists identified functional ortholog of veal2 in humans from HUVECs and named it as VEAL2.

TNF-α Induces Endothelial–Mesenchymal Transition Promoting Stromal Development of Pancreatic Adenocarcinoma

The authors showed that TNF-α strongly induced human endothelial cells to undergo endothelial–mesenchymal transition. They also showed that TNF-α-induced EndMT was reversible. Furthermore, TNF-α treatment of orthotopic mice resulted in an important increase in the stroma, including cancer-associated fibroblasts.

Itacitinib Prevents Xenogeneic GVHD in Humanized Mice

Mice were followed for xenogeneic graft-versus-host disease score and survival. Human T-cell engraftment and as well as human T-cell subtypes were monitored in blood on days 14, 21, and 28 after transplantation.

Nicotinamide N-Methyltransferase in Endothelium Protects against Oxidant Stress-Induced Endothelial Injury

Researchers characterized the functional activity of nicotinamide N-methyltransferase (NNMT) in the endothelium and tested whether NNMT regulated endothelial cell viability.

Efficacious, Safe and Stable Inhibition of Corneal Neovascularization by AAV Vectored Anti-VEGF Therapeutics

To explore more efficacious and long-lasting treatment of corneal neovascularization (CoNV), investigators employed rAAV2 and rAAV8 vectors to mediate KH902 expression via a single intrastromal injection and investigated its anti-angiogenic effects and safety in both alkali burn- and suture-induced CoNV mouse models.

Th17/IL-17 Induces Endothelial Cell Senescence via Activation of NF-κB/p53/RB Signaling Pathway

The proportion of Th17 cells in the spleen and the expression levels of IL-17A, IL-6, and vascular cell adhesion molecule- in mice of different ages were increased with aging.

Impairing Flow-Mediated Endothelial Remodeling Reduces Extravasation of Tumor Cells

The authors aimed at dissecting the cellular and molecular mechanisms driving blood flow-dependent endothelial remodeling. Transcriptomic analysis of endothelial cells revealed that blood flow enhanced VEGFR signaling.

Genetic Engineering of Porcine Endothelial Cell Lines for Evaluation of Human-to-Pig Xenoreactive Immune Responses

A porcine endothelial cell was characterized and immortalized for genetic modification. Five genes including GGTA1, CMAH, β4galNT2, SLA-I α chain, and β2-microglobulin that are responsible for the production of major xenoantigens were sequentially disrupted in immortalized porcine endothelial cells using CRISPR/Cas9 technology.

Selenium Nanoparticles with Various Morphology for Antiangiogenesis through bFGF-Mediated P13K/AKT Signaling Pathways

Selenium nanoparticles could competitively inhibit the binding of bFGF to fibroblast growth factor receptor through direct binding to bFGF, down-regulate the expression of bFGF in HUVEC cells, and significantly reduce the MAPK/Erk and P13K/AKT pathways activation of signaling molecules to regulate HUVEC cell migration and angiogenesis.
