Selective Thrombosis of Tumor for Enhanced Hypoxia-Activated Prodrug Therapy

By encapsulating coagulation-inducing protease thrombin and a hypoxia-activated prodrug tirapazamine into metal–organic framework nanoparticles with a tumor-homing ligand, the obtained nanoplatform selectively activated platelet aggregation at the tumor to induce thrombosis and vascular obstruction therapy by the exposed thrombin.

Fetal Hematopoietic Stem Cell Homing Is Controlled by VEGF Regulating the Integrity and Oxidative Status of the Stromal-Vascular Bone Marrow Niches

By studying physiological homing of fetal hematopoietic stem and progenitor cells (HSPCs), scientists showed the critical requirement of balanced local crosstalk within the skeletal niche for successful HSPC settlement in bone marrow.

The Amot/Integrin Protein Complex Transmits Mechanical Forces Required for Vascular Expansion

Investigators showed that deletion of the scaffold protein Angiomotin (Amot) inhibited migration and expansion of the physiological and pathological vascular network

EPAS1 (Endothelial PAS Domain Protein 1) Orchestrates Transactivation of Endothelial ICAM1 (Intercellular Adhesion Molecule 1) by Small Nucleolar RNA Host Gene 5 (SNHG5 ) to Promote Hypoxic Pulmonary Hypertension

Scientists confirmed that EPAS1-specific inhibitor PT2385 ameliorated HPH features, as demonstrated by right ventricle hypertrophy, right ventricular systolic pressure, and pulmonary vascular remodeling.

Role of Aneuploid Circulating Tumor Cells and CD31+ Circulating Tumor Endothelial Cells in Predicting and Monitoring Anti-angiogenic Therapy Efficacy in Advanced NSCLC

Co-detection and comprehensive phenotypic and karyotypic molecular characterization of aneuploid circulating tumor cells and circulating tumor endothelial cells were conducted on non-small cell lung cancer patients receiving bevacizumab plus chemotherapy.

C1q/Tumor Necrosis Factor-Related Protein-3 Improves Microvascular Endothelial Function in Diabetes through the AMPK/eNOS/NO· Signaling Pathway

Investigators demonstrated that the C1q-like globular domain of C1q/tumor necrosis factor-related protein (CTRP)3, CTRP5, and CTRP9 exerted a vasorelaxant effect on the microvasculature, of which gCTRP3 was the most powerful one.

KLF11 Protects against Venous Thrombosis via Suppressing Tissue Factor Expression

Utilizing stasis-induced murine deep vein thrombosis model and cultured endothelial cells, the authors identified an increase of KLF11 expression under prothrombotic conditions both in vivo and in vitro.

p300/Sp1-Mediated High Expression of p16 Promotes Endothelial Progenitor Cell Senescence Leading to the Occurrence of Chronic Obstructive Pulmonary Disease

Peripheral blood endothelial progenitor cells were isolated from nonsmoking non-chronic obstructive pulmonary disease (COPD), smoking non-COPD, and smoking COPD patients. The expressions of p16, p300, and senescence-related genes were detected by RT-PCR and Western Blot.

Irisin Enhances Angiogenesis of Mesenchymal Stem Cells to Promote Cardiac Function in Myocardial Infarction via PI3k/Akt Activation

Researchers investigated the potential role of irisin in the angiogenesis of bone-marrow mesenchymal stem cells (BMSCs). In vivo, irisin-treated BMSCs were transplanted into an myocardial infarction mouse model.

Putting Function Back in Dysfunction: Endothelial Diseases and Current Therapies in Hematopoietic Stem Cell Transplantation and Cellular Therapies

Scientists summarize the current knowledge about statins and other agents used as endothelial protectants. They propose further studies using statins for prophylaxis and prevention of end-organ damage related to extensive endothelial dysfunction in hematopoietic stem cell transplantation and chimeric antigen receptor-T cells.

HUTCHMED Initiates a Phase Ib/II Trial of Fruquintinib in Combination with Tislelizumab in Advanced Triple Negative Breast Cancer or Advanced Endometrial Cancer

HUTCHMED initiated a Phase Ib/II study of fruquintinib in combination with BeiGene’s tislelizumab in patients with advanced triple negative breast cancer or advanced endometrial cancer in the US. This trial is to explore the potential for the addition of a highly selective vascular endothelial growth factor receptor inhibitor, fruquintinib, to anti-programmed death-1 antibody tislelizumab in inducing activity to immune checkpoint inhibitors.
