    Ancient Viral Molecules Essential for Human Development
    Scientists have shown that certain noncoding RNA molecules, which are genetic remnants of an ancient virus, play a critical role in balancing decisions in early human development–namely pluripotency of early stem cells, a genetic state that enables them to modulate the gamut of human developmental decisions and timing.     The Metabolome Regulates the Epigenetic Landscape during Naive-to-Primed Human Embryonic Stem Cell Transition
    Researchers showed that nicotinamide N-methyltransferase (NNMT) and the metabolic state regulate pluripotency in human embryonic stem cells (hESCs). In naive hESCs, NNMT and its enzymatic product 1-methylnicotinamide are highly upregulated, and NNMT is required for low S-adenosyl methionine levels and the H3K27me3 repressive state.     Parthenogenetic Haploid Embryonic Stem Cells Efficiently Support Mouse Generation by Oocyte Injection
    The authors showed that parthenogenetic (PG)-haploid embryonic stem cells (haESCs) exhibit highly similar gene expression profiles to those of androgenetic-haESCs, and after removal of H19 and Gtl2 differentially methylated DNA regions, DKO-PG-haESCs can efficiently produce semi-cloned pups.     Selection and Dynamics of Embryonic Stem Cell Integration into Early Mouse Embryos
    The process by which pluripotent cells incorporate into host embryos is of interest to investigate cell potency and cell fate decisions. By comparing a heterogeneous mix of undifferentiated and differentiating embryonic stem cell with more homogeneous undifferentiated culture, the authors examined the role of cellular heterogeneity in this process.     Recombinant Xeno-Free Vitronectin Supports Self-Renewal and Pluripotency in Protein Induced Pluripotent Stem Cells
    Investigators tested whether recombinant human matrix proteins can support self-renewal and pluripotency of protein-induced pluripotent stem cells (piPSCs). After long term culture on recombinant human vitronectin in xeno-free conditions, piPSCs retained the expression of pluripotent markers. [Tissue Eng Part C Methods] Abstract

    Generation of Airway Epithelial Cells with Native Characteristics from Mouse Induced Pluripotent Stem Cells
    Scientists generated airway epithelial cells from embryoid bodies by culturing them under serum-free conditions supplemented with Activin and bFGF and by the air-liquid interface method and characterized the induced pluripotent stem cell-derived airway epithelial cells in terms of their gene expression, immunoreactivity, morphology, and function.
