PUBLICATIONS(Ranked by impact factor of the journal)Bioelectrical Domain Walls in Homogeneous Tissues Investigators mapped bioelectrical domain wall motion using all-optical electrophysiology in an engineered cell line and in human iPSC-derived myoblasts. Bioelectrical domain wall migration occurred during embryonic development and during physiological signaling processes in polarized tissues. [Nat Phys] Abstract JMJD3 and UTX Determine Fidelity and Lineage Specification of Human Neural Progenitor Cells Scientists deleted KDM6s (JMJD3 and/or UTX), the H3K27me3 demethylases, in human (h) ESCs and showed that their deletion did not impede neural progenitor cells generation from hESCs. However, KDM6-deficient NPCs exhibited poor proliferation and a failure to differentiate into neurons and glia. [Nat Commun] Full Article A Transcriptome-Wide Antitermination Mechanism Sustaining Identity of Embryonic Stem Cells The authors showed that the RNA-associated protein Srrt/Ars2 sustains ESC identity by preventing premature termination of numerous transcripts at cryptic cleavage/polyadenylation sites in first introns. [Nat Commun] Abstract Cohesin Disrupts Polycomb-Dependent Chromosome Interactions in Embryonic Stem Cells Researchers examined chromosome conformation in ESCs lacking cohesin and found, as in other cell types, that cohesin was required to create topologically associating domains and regulate A/B compartmentalization. However, in the absence of cohesin, they identified a series of long-range chromosomal interactions that persisted. [Cell Rep] Full Article | Graphical Abstract Transcriptional Programming of Human Mechanosensory Neuron Subtypes from Pluripotent Stem Cells Combining NGN2 and BRN3A programming with neural crest patterning, scientists produced two populations of sensory neurons, including a specialized touch receptor neuron subtype. [Cell Rep] Full Article | Graphical Abstract Using serum-, xeno-, and feeder-free stepwise hematopoietic induction protocols, the authors showed that short-term and high-concentration treatment of human PSCs with bone morphogenetic protein 4 (BMP4) strongly promoted early mesoderm induction followed by increased hematopoietic commitment. [Exp Mol Med] Full Article Investigators developed a Mixed Lineage Leukemia (MLL1)-inducible ESC system and showed that induction of wild-type MLL1 during ESC differentiation selectively increased hematopoietic potential from a transitional c-Kit+/Cd41+ population in the embryoid body and also at sites of hematopoiesis in embryos. [Stem Cell Reports] Full Article | Graphical Abstract Researchers generated human PSC lines harboring AAVS1-integrated, inducible and fluorescent dead (d)Cas9-KRAB and dCas9-VPR transgenes to allow for accurate quantification and tracking of cells that expressed both the dCas9 effectors and guide (g)RNAs.
