Metabolic Control of DNA Methylation in Naive Pluripotent Cells

The authors showed that LIF-Stat3 signaling induced genomic hypomethylation via metabolic reconfiguration. Stat3−/− ESCs showed decreased α-ketoglutarate production from glutamine, leading to increased Dnmt3a and Dnmt3b expression and DNA methylation.

Pathogenic LMNA Variants Disrupt Cardiac Lamina-Chromatin Interactions and De-Repress Alternative Fate Genes

Researchers introduced LMNA mutations from individuals with dilated cardiomyopathy into human (h)iPSCs and found that hiPSC-derived cardiomyocytes, in contrast to hepatocytes or adipocytes, exhibited aberrant nuclear morphology and specific disruptions in peripheral chromatin.

Single-Cell Transcriptomic Reveals Molecular Diversity and Developmental Heterogeneity of Human Stem Cell-Derived Oligodendrocyte Lineage Cells

    Investigators describe a time-course single-cell-transcriptomic analysis of developing human stem cell-derived oligodendrocyte-lineage-cells.

3D Bioprinting of High Cell-Density Heterogeneous Tissue Models through Spheroid Fusion within Self-Healing Hydrogels

Scientists bioprinted iPSC-derived cardiac microtissue models with spatially controlled cardiomyocyte and fibroblast cell ratios to replicate the structural and functional features of scarred cardiac tissue that arise following myocardial infarction, including reduced contractility and irregular electrical activity.

Cross-Species Single-Cell Transcriptomic Analysis Reveals Pre-Gastrulation Developmental Differences among Pigs, Monkeys, and Humans

The authors constructed a complete single-cell landscape of early embryonic development of pig, considered one of the best host species for human organ generation, and systematically compared its epiblast development with that of human and monkey.

Generation of Human-Induced Pluripotent Stem Cell-Derived Functional Enterocyte-Like Cells for Pharmacokinetic Studies

Scientists demonstrated the generation of enterocyte-like cells by culturing human iPSC-derived intestinal progenitor cells on a collagen vitrigel membrane.

Cell-to-Cell Adhesion and Neurogenesis in Human Cortical Development: A Study Comparing 2D Monolayers with 3D Organoid Cultures

The authors compared transcriptome and cellular phenotypes between telencephalic organoids and monolayers generated in parallel from three biologically distinct iPSC lines.

Cell Type-Specific Changes in Transcriptomic Profiles of Endothelial Cells, iPSC-Derived Neurons and Astrocytes Cultured on Microfluidic Chips

Investigators analyzed the transcriptomic profiles of co-cultures of human iPSC-derived neurons and rat astrocytes, as well as one-day monocultures of human endothelial cells, cultured on microfluidic chips.

Cell-Based Strategies for IVD Repair: Clinical Progress and Translational Obstacles

Scientists discuss current approaches to disc regeneration, with a particular focus on cell-based therapeutic strategies, including ongoing challenges, and attempts to provide a framework to interpret current data and guide future investigational studies.

Chromatin Accessibility Dynamics during Cell Fate Reprogramming

Investigators discuss recent findings linking chromatin dynamics to cell fate control. Specifically, chromatin undergoes a binary off/on switch during iPSC reprogramming, closing and opening loci occupied by somatic and pluripotency transcription factors, respectively.
