APOE2 Mitigates Disease-Related Phenotypes in an Isogenic hiPSC-Based Model of Alzheimer’s Disease

Scientists utilized an isogenic human induced pluripotent stem cell (hiPSC)-based system to demonstrate that conversion of APOE3 to APOE2 greatly reduced the production of amyloid-beta peptides in hiPSC-derived neural cultures.

Centrosome‐Mediated Microtubule Remodeling During Axon Formation in Human iPSC‐Derived Neurons

The authors demonstrated that centrosomes played an essential role in controlling axon formation in hiPSC‐derived neurons.

Establishment of Bovine Expanded Potential Stem Cells

Investigators explored the expanded potential stem cell (EPSC) technology for mammalian species refractory to the standard ESC approaches and report the successful establishment of bovine EPSCs from preimplantation embryos of both wild-type and somatic cell nuclear transfer.

Efficient Conversion of Human Induced Pluripotent Stem Cells Into Microglia by Defined Transcription Factors

The authors report a novel approach that produces, within ten days, the differentiation of human induced pluripotent stem cells into microglia by forced expression of both SPI1 and CEBPA.

Fluid Shear Stress Promotes Embryonic Stem Cell Pluripotency via Interplay between β‐catenin and Vinculin in Bioreactor Culture

Researchers investigated how murine ESCs sense fluid shear stress and modulate a non‐canonical Wnt signaling response to promote pluripotency.

Method for Selective Ablation of Undifferentiated Human Pluripotent Stem Cell Populations for Cell-Based Therapies

Scientists determined an appropriate in vitro doxorubicin dose that (a) eliminates residual undifferentiated stem cells before cell injection to prevent teratoma formation after cell transplantation and (b) does not cause cardiotoxicity in ESC-derived cardiomyocytes as demonstrated through contractility analysis, electrophysiology, topoisomerase activity assay, and quantification of reactive oxygen species generation.

Gene and Protein Expression in Human Megakaryocytes Derived From Induced Pluripotent Stem Cells

iPSCs were reprogrammed from peripheral blood mononuclear cells and megakaryocytes were derived from the iPSCs in 194 healthy European American and African American subjects. mRNA was isolated and gene expression measured by RNA sequencing. Protein expression was measured in 62 of the subjects using mass spectrometry.

    Abstract

Physiological and Pharmacological Stimulation for In Vitro Maturation of Substrate Metabolism in Human Induced Pluripotent Stem Cell-Derived Cardiomyocytes

Researchers investigated substrate metabolism in hiPSC-CMs grown as a monolayer and in 3D, in porous collagen-derived scaffolds and in engineered heart tissue, by measuring rates of glycolysis and glucose and fatty acid oxidation, and changes in gene expression and mitochondrial oxygen consumption.

Defining Optimal Enzyme and Matrix Combination for Replating of Human Induced Pluripotent Stem Cell-Derived Cardiomyocytes at Different Levels of Maturity

Investigators revealed the selection criteria of the dissociation approach and coating matrix for replating iPSC-cardiomyocytes based on the maturity and the requirements of further applications.
