    Label-Free Imaging for Quality Control of Cardiomyocyte Differentiation

    Investigators developed a non-invasive label-free live cell imaging platform to predict the efficiency of human PSC (hPSC) differentiation into cardiomyocytes. Autofluorescence imaging of metabolic co-enzymes was performed under varying differentiation conditions across five hPSC lines.

    LIN28 Coordinately Promotes Nucleolar/Ribosomal Functions and Represses the 2C-Like Transcriptional Program in Pluripotent Stem Cells

    Researchers showed that LIN28 displayed dynamic temporal and spatial expression during murine embryo development. In cultured PSCs, loss of LIN28 led to nucleolar stress and activation of a 2-cell/4-cell-like transcriptional program characterized by the expression of endogenous retrovirus genes.

    Arrest of WNT/β-Catenin Signaling Enables the Transition from Pluripotent to Differentiated Germ Cells in Mouse Ovaries

    The authors showed in mouse embryonic ovaries that germ cell–intrinsic β-catenin activity maintained pluripotency and that its repression was essential to allow differentiation and meiosis entry in a timely manner.

    Establishment of a KCNQ1 Homozygous Knockout Human Embryonic Stem Cell Line by Episomal Vector-Based CRISPR/Cas9 System

    Investigators generated a homozygous KCNQ1 knockout human ESC line using episomal vector-based CRISPR/Cas9 system. This generated cell line presented typical stem cells colony morphology, maintained high pluripotency and normal karyotype, also was able to differentiate into all three germ layers in vivo.

    Establishment of Two Homozygous CRISPR Interference (CRISPRi) Knock-In Human Induced Pluripotent Stem Cell (hiPSC) Lines for Titratable Endogenous Gene Repression

    The authors inserted CRISPRi cassette into the AAVS1 locus of hiPSC lines, which resulted in homozygous knock-in with an otherwise unaltered genome. Expression of dCas9KRAB protein, pluripotency and the ability to differentiate into all three embryonic germ layers were validated.

    Recapitulative Hematopoietic Development of Human Pluripotent Stem Cells in the Absence of Exogenous Hematopoietic Cytokines

    Scientists showed that endogenous stimuli and VEGF were sufficient to induce robust human PSC-derived hematopoiesis, intensive generation of hematopoietic progenitors, maturation of blood cells and the emergence of definitive precursor cells including those that were phenotypically identical to early human embryonic hematopoietic stem cells.
