    TOP STORY

Efficient Derivation of Human Trophoblast Stem Cells from Primed Pluripotent Stem Cells

Scientists reported the successful generation of trophoblast stem cells (TSCs) from primed human PSCs (hPSCs) and showed that BMP4 substantially enhanced this process. TSCs derived from primed hPSCs were similar to blastocyst-derived human TSCs in terms of morphology, proliferation, differentiation potential, and gene expression.

Chd1 Protects Genome Integrity at Promoters to Sustain Hypertranscription in Embryonic Stem Cells

Researchers reported a novel role for Chd1 in protecting genome integrity at promoter regions by preventing DNA double-stranded break accumulation in ESCs.

Topological Isolation of Developmental Regulators in Mammalian Genomes

Investigators generated deeply sequenced Hi-C data for human PSCs that allowed them to identify CTCF loop domains that had highly conserved boundary CTCF sites and showed a notable enrichment of individual developmental regulators.

Genetically Variant Human Pluripotent Stem Cells Selectively Eliminate Wild-Type Counterparts through YAP-Mediated Cell Competition

The authors demonstrated that human PSC fate was coupled to mechanical cues imposed by neighboring cells and revealed that hijacking this mechanism allowed variants to achieve clonal dominance in cultures.

iPSC-Derived Neoantigen-Specific Cytotoxic T-Lymphocyte Therapy for Ewing Sarcoma

Researchers examined the antitumor effects of EWS/FLI1-rejuvenated cytotoxic T lymphocytes against Ewing sarcoma. The altered amino acid sequence at the break/fusion point of EWS/FLI1, when presented as a neoantigen, evoked an immune response that targeted EWS/FLI1+ sarcoma.

Perichondrium-Inspired Permeable Nanofibrous Tube Well Promoting Differentiation of hiPSC-Derived Pellet toward Hyaline-Like Cartilage Pellet

The regulated spatially homogeneous chondrogenic microenvironment around the human iPSC-derived pellet in the perichondrium-inspired permeable nanofibrous tube well remarkably improved the quality of the differentiated pellet toward a more hyaline-like cartilage pellet.

Selective Deletion of Human Leukocyte Antigens Protects Stem Cell-Derived Islets from Immune Rejection

Scientists used genetic engineering to create human PSCs in which the majority of the polymorphic human leukocyte antigens, the main drivers of allogeneic rejection, were deleted.

An In Vitro Stem Cell Model of Human Epiblast and Yolk Sac Interaction

Investigators established conditions to differentiate human PSCs into yolk sac-like cells that resembled the post-implantation human hypoblast molecularly and functionally.

The Combined Action of Esrrb and Nr5a2 Is Essential for Murine Naïve Pluripotency.

Researchers reported that the conjunct activity of two orphan nuclear receptors, Esrrb and Nr5a2, paralleled the importance of that of Oct4 and Sox2 in naïve ESCs.

Rapid Target Validation in a Cas9-Inducible hiPSC Derived Kidney Model

The authors presented a methodology for direct differentiation of the CRISPR-targeted cell pools, using a doxycycline-inducible Cas9 expressing human iPSC line for high efficiency editing to eliminate the laborious clonal line generation steps.

Implantation of Human-Induced Pluripotent Stem Cell-Derived Cartilage in Bone Defects of Mice

Scientists prepared cartilage from human iPSCs (hiPS-Cart) in a scaffoldless manner and implanted hiPS-Cart into 3.5 mm large defects created in the femurs of immunodeficient mice to examine the repair capacity.
