    ATF6 Is Essential for Human Cone Photoreceptor Development

    Scientists generated retinal organoids from achromatopsia patient iPSCs carrying ATF6 disease variants and from gene-edited ATF6 null human ESCs, and found that achromatopsia patient and ATF6 null retinal organoids failed to form cone structures concomitant with loss of cone phototransduction gene expression, while rod photoreceptors developed normally.

    The Conserved Fertility Factor SPACA4/Bouncer Has Divergent Modes of Action in Vertebrate Fertilization

    Researchers showed that Bouncer’s homolog in mammals, Sperm Acrosome Associated 4 (SPACA4), was required for efficient fertilization in mice. Male knockout mice were severely subfertile, and sperm lacking SPACA4 failed to fertilize wild-type eggs in vitro.

    iPSC Culture Expansion Selects against Putatively Actionable Mutations in the Mitochondrial Genome

    Sequencing of mitochondrial DNA (mtDNA) of 26 iPSC clones did not reveal evidence for de novo mutagenesis, or for any selection processes during reprogramming or differentiation. Culture expansion, however, selected against putatively actionable mtDNA mutations.

    Novel Population of Human Monocyte and Osteoclast Progenitors from Pluripotent Stem Cells and Peripheral Blood
