Targeting PAK4 to Reprogram the Vascular Microenvironment and Improve CAR-T Immunotherapy for Glioblastoma

PAK4 knockout induced adhesion protein re-expression in endothelial cells, reduced vascular abnormalities, improved T cell infiltration and inhibits glioblastoma growth in mice.

Tumor-Associated Macrophages Drive Stromal Cell-Dependent Collagen Crosslinking and Stiffening to Promote Breast Cancer Aggression

Scientists profiled lysyl hydroxylase-mediated and lysyl oxidase-mediated collagen crosslinks and quantified the greatest abundance of total and complex collagen crosslinks in aggressive human breast cancer subtypes with the stiffest stroma.

Derivation of Intermediate Pluripotent Stem Cells Amenable to Primordial Germ Cell Specification

By modulating fibroblast growth factor, transforming growth factor β, and WNT pathways, the authors derived pluripotent stem cells from mice, horses, and humans that were permissive for direct primordial germ cell-like cell induction in vitro and were capable of contributing to intra- or inter-species chimeras in vivo.

Differentiated Daughter Cells Regulate Stem Cell Proliferation and Fate through Intra-Tissue Tension

Using two distinct mouse lines, scientists found that increasing contractility of differentiated cells resulted in non-cell-autonomous hyperproliferation of stem cells and prevented their commitment to a hair follicle lineage.

Composition, Spatial Characteristics, and Prognostic Significance of Myeloid Cell Infiltration in Pancreatic Cancer

To generate spatially-resolved tumor and immune cell data at single cell resolution, investigators developed two quantitative multiplex immunofluorescence assays to interrogate myeloid cells and macrophages in the pancreatic ductal carcinoma tumor microenvironment.

The Microfluidic Environment Reveals a Hidden Role of Self-Organizing Extracellular Matrix in Hepatic Commitment and Organoid Formation of hiPSCs

Hepatic progenitor cells either derived in microfluidics or exposed to exogenous ECM stimuli showed a significantly higher potential of forming hepatic organoids that could be rapidly expanded for several passages and further differentiated into functional hepatocytes.

Macrophage-Derived Small Extracellular Vesicles Promote Biomimetic Mineralized Collagen-Mediated Endogenous Bone Regeneration

Researchers evaluated the potential role of macrophage-derived small extracellular vesicles (sEVs) in biomimetic mineralized collagen-mediated endogenous bone regeneration in vivo and in vitro. Macrophage-derived sEVs were involved in intrafibrillarly mineralized collagen (IMC)-mediated endogenous bone regeneration in vivo, and IMC-sEVs facilitated mesenchymal stem cell osteogenesis through the Smad/1/5/9 pathway.

Antagonistic Effects of Activin A and Tnf-α on the Activation of L929 Fibroblast Cells via Smad3-Independent Signaling

Scientists revealed that TNF-α promoted the release of nitric oxide and IL-6 by L929 fibroblast cells, but co-treatment with activin A attenuated these effects. In contrast, activin A induced cell migration and increased the production of tissue fibrosis-related TGF-β1 and fibronectin, while TNF-α inhibited these function changes of L929 cells induced by activin A.

Systems Biology Analysis of Osteogenic Differentiation Behavior by Canine Mesenchymal Stem Cells Derived from Bone Marrow and Dental Pulp

The authors illustrated that canine dental pulp stem cells contained superior osteogenicity comparing with canine bone marrow-derived mesenchymal stem cells regarding alkaline phosphatase activity, matrix mineralization, and osteogenic marker expression.

Insight into Cisplatin-Resistance Signaling of W1 Ovarian Cancer Cells Emerges mTOR and HSP27 as Targets for Sensitization Strategies

The microenvironment possesses a strong impact on the tumor chemoresistance when cells bind to components of the ECM. Scientists elucidated the signaling pathways of cisplatin resistance in W1 ovarian cancer cells binding to collagen type 1 and signaling interference with constitutive cisplatin resistance in W1CR cells to discover the targets for sensitization.

    INDUSTRY AND POLICY NEWS

Turnstone Biologics Announces First Patients Enrolled in Phase I/II Clinical Trial Evaluating RIVAL-01 for the Treatment of Solid Tumors

Turnstone Biologics Corp. has enrolled the first patients in a Phase I/IIa clinical trial of its RIVAL-01/TAK-605 candidate in patients with solid tumors, conducted in collaboration with Takeda Pharmaceutical Company Limited. RIVAL-01 consists of Turnstone’s proprietary oncolytic vaccinia virus backbone encoding transgenes for Flt3 ligand, anti-CTLA-4 antibody, and IL-12 cytokine, purposefully designed to work together to drive immune activity and re-program the microenvironment to be best suited for tumor eradication.
