Xenogenization of Tumor Cells by Fusogenic Exosomes in Tumor Microenvironment Ignites and Propagates Antitumor Immunity

Researchers suggested a previously unidentified method for xenogenizing tumors by using fusogenic exosomes to introduce fusogenic viral antigens onto the tumor cell surface.

Esophageal Extracellular Matrix Hydrogel Mitigates Metaplastic Change in a Dog Model of Barrett’s Esophagus

Dogs with Barrett’s esophagus were treated orally with esophageal mucosa ECM (eECM) hydrogel and omeprazole or omeprazole alone for 30 days. eECM treatment resolved esophagitis, reverted metaplasia to a normal, squamous epithelium in four of six animals.

Self-Assembled Dodecyl Group-Modified Gelatin Microparticle-Based Hydrogels with Angiogenic Properties

The authors designed a novel angiogenic microparticle (C12-MP) comprising Alaska pollock-derived gelatin (ApGltn) modified with a dodecyl group (C12-ApGltn) to stimulate endogenous vascular endothelial growth factor secretion.

BMP-1 Disrupts Cell Adhesion and Enhances TGF-β Activation through Cleavage of the Matricellular Protein Thrombospondin-1

Bone morphogenetic protein 1 (BMP-1) overexpression induced marked phenotypic changes in two human cell lines. Scientists investigated how BMP-1 simultaneously affected cell-matrix interactions and growth factor activity in these cells.

    Abstract

IQGAP1 Promotes Anoikis Resistance and Metastasis through Rac1-Dependent ROS Accumulation and Activation of Src/FAK Signaling in Hepatocellular Carcinoma

IQGAP1 expression was detected by immunohistochemistry, real-time PCR and immunoblot analysis. Lentiviral-mediated stable upregulation or knockdown of IGAQP1, immunoprecipitation, etc. were used in function and mechanism study.

    Abstract

Cancer-Associated Fibroblasts Mediate Cancer Progression and Remodel the Tumoroid Stroma

Cancer-associated fibroblasts enhanced the distance and surface area of the invasive cancer mass whilst inhibiting vascular-like network formation. These processes correlated with the upregulation of hepatocyte growth factor, metallopeptidase inhibitor 1 and fibulin-5.

    Abstract

EPS8 Phosphorylation by Src Modulates Its Oncogenic Functions

Plasmids expressing scaffolding protein, EPS8 Src-mediated phosphorylation site mutants were expressed in cells containing a normal endogenous level of EPS8. In addition, cells were treated with dasatinib to inhibit Src activity.

    Abstract

Cell Derived Matrix Fibulin-1 Associates with Epidermal Growth Factor Receptor to Inhibit Its Activation, Localization and Function in Lung Cancer Calu-1 Cells

To test the possible role Fibulin-1 isoforms could have in regulating Epidermal Growth Factor Receptor signaling and function in lung cancer, scientists performed siRNA mediated knockdown of FBLN1C and FBLN1D in non-small cell lung cancer Calu-1 cells.

Genetic Reduction of the Extracellular Matrix Protein Versican Attenuates Inflammatory Cell Infiltration and Improves Contractile Function in Dystrophic mdx Diaphragm Muscles

Versican reduction improved the ex vivo strength and endurance of diaphragm muscle strips. These changes in diaphragm contractile properties in mdx-hdf mice were associated with decreased monocyte and macrophage infiltration and a reduction in the proportion of fibres expressing the slow type I myosin heavy chain isoform

Laminin-511-E8 Promotes Efficient In Vitro Expansion of Human Limbal Melanocytes

Scientists established a protocol for the efficient isolation and cultivation of pure populations of human limbal melanocytes, which could be expanded at high yield by using recombinant laminin-511-E8 as culture substrate.

    INDUSTRY AND POLICY NEWS

ProLynx Announces SBIR Grant Award to Develop Long-Acting Parathyroid Hormone for Hypo-Parathyroidism

ProLynx announced it was awarded a Small Business Innovation Research (SBIR) grant from the NIH. The grant was conferred for the development of a long acting parathyroid hormone – PTH(1-34) – that can be administered once-weekly for treatment of hypo-parathyroidism. To achieve this, ProLynx will employ its platform half-life extension technology using its hydrogel microsphere carriers.
