    TOP STORY

Blood Stem Cells Emerge from Aortic Endothelium by a Novel Type of Cell Transition
Using non-invasive, high-resolution imaging of live zebrafish embryos, researchers show that hematopoietic stem cells emerge directly from the aortic floor, through a stereotyped process that does not involve cell division but a strong bending then egress of single endothelial cells from the aortic ventral wall into the sub-aortic space, and their concomitant transformation into hematopoietic cells. LABORATORY RESEARCH

In Vivo Imaging of Hematopoietic Cells Emerging from the Mouse Aortic Endothelium
The dynamic de novo emergence of phenotypically defined hematopoietic stem cells (Sca1+, c-kit+, CD41+) directly from ventral aortic hemogenic endothelial cells was illustrated. [Nature]

Impact of Interactions of Cellular Components of the Bone Marrow Microenvironment on Hematopoietic Stem and Progenitor Cell Function
Researchers examined the impact of hematopoietic niche elements on hematopoietic stem and progenitor function by analyzing the combined effect of osteoblasts and stromal cells on Lineage- Sca1+ CD117+ cells. [Blood]

The Midregion, Nuclear Localization Sequence, and C Terminus of PTHrP Regulate Skeletal Development, Hematopoiesis, and Survival in Mice
The results establish that the midregion, nuclear localization sequence, and C terminus of parathyroid hormone-related protein (PTHrP) are crucial for the commitment of osteogenic and hematopoietic precursors to their lineages, and for survival, and many of the effects of PTHrP on development are not mediated by its N terminus. [FASEB J]

Dasatinib Inhibits the Growth of Molecularly Heterogeneous Myeloid Leukemias
Because Src kinases contribute to multiple blood cell functions by triggering a variety of signaling pathways, researchers hypothesized that their molecular targeting might lead to growth inhibition in acute myeloid leukemia. [Clin Cancer Res]

Hematopoietic Stem Cells in Coculture with Mesenchymal Stromal Cells – Modelling the Niche Compartments In Vitro
Here researchers investigated in detail the spatial relationship between hematopoietic stem cells and mesenchymal stromal cells during ex-vivo expansion. [Haematologica]

Role of Chk1 in the Differentiation Program of Hematopoietic Stem Cells
Hematopoietic stem cells isolated from umbilical cord blood were treated with ionizing radiation (IR) and sensitivity and IR induced checkpoints activation were investigated. [Cell Mol Life Sci]

Comparative Transfection of DNA into Primary and Transformed Mammalian Cells from Different Lineages
Studies were undertaken to evaluate and compare the transfection efficacy of several chemical reagents to that of the electroporation/nucleofection system using both adherent cells (primary and transformed airway epithelial cells and primary fibroblasts as well embryonic stem cells) and cells in suspension (primary hematopoietic stem/progenitor cells and lymphoblasts). [ BMC Biotechnol]

Genomic Approaches Uncover Increasing Complexities in the Regulatory Landscape at the Human SCL (TAL1) Locus
To elaborate further the DNA elements which control the regulation of the SCL (TAL1) transcription factor, researchers used genomic tiling microarrays covering 256 kb of the human SCL locus to perform a concerted analysis of chromatin structure and binding of regulatory proteins in human hematopoietic cell lines. [PLoS One]

    CLINICAL RESEARCH

Sustained Molecular Response With Interferon Alfa Maintenance After Induction Therapy With Imatinib Plus Interferon Alfa in Patients With Chronic Myeloid Leukemia
Treatment with interferon alfa-2a (IFN) enables discontinuation of imatinib in most patients after prior imatinib/IFN combination therapy and may result in improved molecular response. [J Clin Oncol]

Clinical Characteristics and Outcomes of Adults with Philadelphia Chromosome Positive and/or Bcr-Abl Positive Acute Lymphoblastic Leukemia: a Single Center Study from China
The results of the study indicated that introducing imatinib into treatment of patients with Ph-positive acute lymphoblastic leukemia could improve complete remission rate and survival, especially as early as possible, and allogenic stem cell transplantation was still the first choice for these patients if they had suitable donors. [Leuk Lymphoma]

Outcome of Allogeneic Bone Marrow Transplantation from Unrelated Donors for Adult Philadelphia Chromosome-Negative Acute Lymphocytic Leukemia in First Complete-Remission
The study suggested that unrelated allogeneic stem cell transplantation (allo-SCT) could improve leukemia-free survival of patients with a potential graft-versus-leukemia effect. [Int J Hematol]
