PUBLICATIONS(Ranked by impact factor of the journal)PTPσ Inhibitors Promote Hematopoietic Stem Cell Regeneration Researchers describe small molecule inhibitors of protein tyrosine phosphatase-sigma (PTPσ) that promote HSC regeneration in vivo. Systemic administration of the PTPσ inhibitor, DJ001, or its analog, to irradiated mice promoted HSC regeneration, accelerated hematologic recovery, and improved survival. [Nat Commun] Full Article Investigators identified the actions of Runx1 and Pu.1 itself at the Pu.1 gene upstream regulatory element as major regulators of Pu.1 expression in burst-forming unit erythrocytes. [Proc Natl Acad Sci USA] Abstract The authors tested the HDAd-CRISPR + HDAd-donor vector system in AAVS1 transgenic mice using a standard ex vivo HSC gene therapy approach as well as in a new in vivo HSC transduction approach that involved HSC mobilization and intravenous HDAd5/35++ injections. [Mol Ther] Abstract | Graphical Abstract m6A RNA Methylation Maintains Hematopoietic Stem Cell Identity and Symmetric Commitment Using single-cell RNA sequencing in combination with transcriptomic profiling of hematopoietic stem and progenitor cells from control and m6A methyltransferase Mettl3 conditional knockout mice, scientists found that m6A-deficient HSCs failed to symmetrically differentiate. [Cell Rep] Full Article | Graphical Abstract TCR Sequencing Reveals the Distinct Development of Fetal and Adult Human Vγ9Vδ2 T Cells The authors analyzed the CDR3 of the TCR of human blood and thymic Vγ9Vδ2 T cells from fetal until adult life. They first identified key differences in the CDR3 repertoire of fetal and adult blood Vγ9Vδ2 T cells, including in CDR3 features important for phosphoantigen reactivity. [J Immunol] Abstract Investigators report the effects of graphene quantum dots on the transcriptome of CD34+ HSCs after an incubation time of 36 hours. [Sci Rep] Full Article Bmi1 deletion promoted the adipogenic differentiation of leptin receptor+ stromal cells and caused progressive fatty changes in the bone marrow (BM) of limb bones with age, resulting in reductions in the numbers of HSCs and progenitors in BM and enhanced extramedullary hematopoiesis. [Exp Hematol] Abstract CLINICAL RESEARCHThe outcomes of 1234 adult patients with ALL in first complete remission who underwent allogeneic stem cell transplantation between 2007 and 2016 were analyzed. Comparison was done between haploidentical donor, matched unrelated donor, and mismatched unrelated donor. REVIEWS3D Chromosomal Landscapes in Hematopoiesis and Immunity The authors summarize the current principles of 3D chromatin organization, how the integrity of the 3D genome governs immune cell development and malignant transformation, as well as how underlying (epi-)genetic drivers of 3D chromatin alterations might act as potential novel therapeutic targets for hematological malignancies. POLICY NEWSBrazil’s Budget Cuts Threaten More Than 80,000 Science Scholarships Brazil’s main science-funding agency will have to suspend more than 80,000 scholarships to postdoctoral researchers and graduate and undergraduate students starting in September unless it receives additional cash from the government. [Nature News] Editorial The Push to Replace Journal Supplements with Repositories Broken links, clunky formats, and outdated platforms have both authors and publishers turning to alternative solutions.
