TOP STORYThe Bone Marrow Protects and Optimizes Immunological Memory during Dietary Restriction Scientists showed that memory T cells collapsed in secondary lymphoid organs in the context of dietary restriction but dramatically accumulated within the bone marrow, where they adopted a state associated with energy conservation. PUBLICATIONS(Ranked by impact factor of the journal)Precision Mouse Models with Expanded Tropism for Human Pathogens Researchers showed that implantation of human lung tissue, which contains up to 40 cell types, including nonhematopoietic cells, into immunodeficient mice (lung-only mice) resulted in the development of a highly vascularized lung implant. [Nat Biotechnol] Abstract Investigators found that hCINAP was involved in the repair of double-stranded DNA breaks (DSB) and that its expression correlated with acute myeloid leukemia prognosis. Following DSB, hCINAP was recruited to damage sites where it promoted SENP3-dependent deSUMOylation of NPM1. [Nat Commun] Full Article The authors generated isogenic TP53-knockout versions of the euploid acute myeloid leukemia cell line EEB to investigate the impact of TP53 on karyotype stability. They showed that chromosome abnormalities spontaneously arose in TP53-deficient cells. [Leukemia] Abstract Scientists analyzed the relationship between chemokine receptor 4 (CXCR4) expression and the FLT3-ITD mutation in 466 patients with de novo AML to clarify the effect of FLT3-ITD mutations on CXCR4 expression in patients with AML. [Sci Rep] Full Article By using shRNAi to target GATA2 in acute myeloid leukemia (AML) cell lines, and an AML cell line expressing normal levels of GATA2, the authors found that inhibition of GATA2 caused attenuated cell proliferation and enhanced apoptosis exclusively in AML cell lines that overexpressed GATA2. [Sci Rep] Full Article Researchers comprehensively analyzed the CD34+ compartment in G-CSF-stimulated peripheral blood stem cell grafts of allogeneic donors, in bone marrow (BM) samples of the respective recipients four weeks after allogeneic HSC transplantation, and in BM samples of healthy donors. [Bone Marrow Transplant] Abstract CLINICAL RESEARCHHuman Herpesvirus 6B and Lower Respiratory Tract Disease after Hematopoietic Cell Transplantation Investigators tested bronchoalveolar lavage (BAL) fluid for human herpesvirus 6B DNA using polymerase chain reaction in allogeneic hematopoietic cell transplantation recipients who underwent a BAL for evaluation of lower respiratory tract disease from 1992 to 2015. [J Clin Oncol] Abstract Researchers performed a retrospective multicenter analysis of 113 patients with refractory/relapsed classic Hodgkin lymphoma who had undergone allogeneic stem cell transplant in Argentina. INDUSTRY NEWSSigilon Therapeutics Receives Orphan Drug Designation for SIG-001 for the Treatment of Hemophilia A Sigilon Therapeutics announced it has received Orphan Drug Designation for SIG-001, an investigational therapy for hemophilia A that leverages Sigilon’s Shielded Living Therapeuticsâ„¢ platform to implant cells engineered to produce stable blood plasma levels of factor VIII, a crucial clotting protein. [Sigilon Therapeutics, Inc.] Press Release GSK Announces Positive Headline Results from the Pivotal DREAMM-2 Study for Multiple Myeloma GlaxoSmithKline plc announced positive headline results from the pivotal DREAMM-2 open-label, randomized study of two doses of belantamab mafodotin. The 196 patients in the trial had relapsed multiple myeloma, were refractory to an immunomodulatory drug, a proteasome inhibitor, and to treatment with an anti-CD38 antibody. [GlaxoSmithKline plc] Press Release Rocket Pharmaceuticals, Inc. announced clearance from the Spanish Agency for Medicines and Health Products to commence enrollment in the FANCOLEN-II Phase II registration-enabling study of RP-L102 for Fanconi anemia in Spain. [Rocket Pharmaceuticals, Inc.] Press Release Genmab Announces Approval of DARZALEX® (Daratumumab) in Frontline Multiple Myeloma in Japan Genmab A/S announced that the Ministry of Health, Labor and Welfare in Japan has approved the use of daratumumab in combination with bortezomib, melphalan and prednisone for the treatment of patients with newly diagnosed multiple myeloma who are ineligible for autologous stem cell transplant. [Genmab A/S] Press Release IU School of Medicine Researcher Hal Broxmeyer Awarded IU President’s Medal In recognition of his impact and influence, Indiana University (IU) President Michael A. McRobbie presented Hal Broxmeyer with the President’s Medal for Excellence, a rare honor which is awarded to individuals at IU for “outstanding academic, artistic or professional accomplishments or to individuals for exceptional service to the university.” [Indiana University] Press Release | |

POLICY NEWSMemo to Italy’s President: Your Researchers Need You Italy’s coalition government ended abruptly, when the nationalist Lega party of deputy prime minister Matteo Salvini announced that it was walking away from its turbulent coalition with the anti-establishment M5S party, known as the Five Star Movement. The collapse is of great concern: a much-delayed funding increase is now on hold, and the political uncertainty adds further threat. [Nature News] Editorial NSF Graduate Fellowships Disproportionately Go to Students at a Few Top Schools As this year’s eligible students begin to labor over their applications, it’s worth asking why certain schools come away with dozens of winners each year—and what that means about the academy at large.
