TOP STORYAlterations in Chromatin at Antigen Receptor Loci Define Lineage Progression during B Lymphopoiesis Using the micrococcal nuclease-based chromatin accessibility assay with low-cell count input, researchers profiled both small-scale and large-scale changes in chromatin accessibility and nucleosome occupancy in primary cells during lymphoid development, tracking the changes as different antigen receptor loci became primed for recombination. PUBLICATIONS(Ranked by impact factor of the journal)G-CSF Directly Acts on Mouse Lymphoid-Biased but not Myeloid-Biased Hematopoietic Stem Cells Scientists performed serum-free single-cell culture and competitive repopulation with cultured cells. Single highly purified hematopoietic stem cells and hematopoietic progenitor cells were cultured with stem cell factor (SCF), SCF + granulocyte colony-stimulating factor (G-CSF), SCF + granulocyte/macrophage-CSF, or SCF + thrombopoietin for seven days. [Haematologica] Abstract To genetically model the ribosomal defect identified in Diamond-Blackfan anemia (DBA), researchers generated a rpl18 mutant line in zebrafish, using CRISPR/Cas9 system. Molecular characterization of this mutant line demonstrated that Rpl18 deficiency mirrored the erythroid defects of DBA, namely a lack of mature red blood cells. [Cell Death Dis] Full Article Investigators examined whether the use of low concentration of transforming growth factor β1 would be beneficial for increasing red blood cell production by stimulating hematopoietic stem cell growth and also supporting erythroid differentiation. [Stem Cell Res Ther] Full Article The role of Cxxc5 in myelopoiesis was studied using overexpression and short hairpin RNA‐mediated knockdown in mouse early stem and progenitor cells defined as Lineage− Sca‐1+c‐Kit+ cells. [J Leukoc Biol] Abstract Zika virus RNA replication, protein translation and infectious particle formation in hematopoietic cell lines as well as primary CD34+ HSPCs and ex vivo differentiated erythroid and megakaryocytic cells was monitored using qRT‐PCR, FACS, immunofluorescence analysis and infectivity assays. [Transfusion] Full Article Inhibition of JAK2 Suppresses Myelopoiesis and Atherosclerosis in Apoe−/− Mice TG101348 selectively reversed neutrophilia, monocytosis, hematopoietic stem and progenitor cell (HSPC), and granulocyte-macrophage progenitor expansion in Apoe−/− mice with decreased cellular phosphorylated STAT5 and ERK1/2 and reduced cell cycling and BrdU incorporation in HSPCs, indicating inhibition of JAK/STAT signaling and cell proliferation. [Cardiovasc Drugs Ther] Full Article The authors deleted a conserved enhancer driving pan-endothelial expression of the zebrafish gata2a and showed that Gata2a was required for hemogenic endothelium programming by regulating expression of runx1 and of the second Gata2 orthologue, gata2b. [Commun Biol] Full Article CLINICAL RESEARCHThis study was performed through the Center for International Blood and Marrow Transplant Research (CIBMTR) database. Investigators retrospectively reviewed all patients reported to the CIBMTR registry from 2002-2014 who underwent HCT for CML and were alive 30 days post relapse. REVIEWSInvestigators describe the current role of autologous and allogeneic hematopoietic stem cell transplantation (HCT) in the treatment of patients with acute leukemias, aggressive B‐cell lymphomas, and multiple myeloma and discuss how novel targeted therapies and immunotherapies have changed the potential need, timing, and goal of HCT in patients with these diseases. [Cancer] Abstract The authors summarize how dysregulation in the expression of genes related to self‐renewal, proliferation, and differentiation of hematopoietic stem cells caused by changes in epigenetic modifications and transcriptional networks lead to clonal expansion and leukemic transformation. POLICY NEWSWhite House Formally Invites Public Comment on Open-Access Policies The White House issued a notice seeking comment on its effort to enhance public access to federally funded research. It’s an old idea creating new controversy. White House Office of Science and Technology Policy Director Kelvin Droegemeier is pushing back against publishers that in December said the administration was quietly pursuing an executive order to require immediate free distribution of taxpayer funded research. [ScienceInsider] Editorial Has the UK Science Minister Been Demoted? A government reshuffle in the United Kingdom has born another new science minister – a position that has seen a revolving door of appointees, firings and resignations in recent years. The latest appointment also comes with an apparent political demotion for the role, which expert observers say is in conflict with a government that appears to have put research and innovation at the heart of its agenda.
