    Immunogenomic Landscape of Hematological Malignancies

Researchers integrated over 8,000 transcriptomes and 2,000 samples with multilevel genomics of hematological cancers to investigate how immunological features are linked to cancer subtypes, genetic and epigenetic alterations, and patient survival, and validated key findings experimentally.

Endogenous Retroviruses Are a Source of Enhancers with Oncogenic Potential in Acute Myeloid Leukemia

The authors identified six endogenous retrovirus families with acute myeloid leukemia (AML)-associated enhancer chromatin signatures that were enriched in binding of key regulators of hematopoiesis and AML pathogenesis.

Integrins Modulate Extracellular Matrix Organization to Control Cell Signaling during Hematopoiesis

Scientists showed that integrins play a key role during fly hematopoiesis in regulating cell signals that control the behavior of hematopoietic progenitors. Integrins could regulate hematopoiesis directly, via focal adhesion kinase signaling, and indirectly, by directing ECM assembly and/or maintenance.

MYSM1 Maintains Ribosomal Protein Gene Expression in Hematopoietic Stem Cells to Prevent Hematopoietic Dysfunction

Investigators describe the transcriptome of Mysm1-deficient mouse hematopoietic stem cells and identified MYSM1 genome-wide DNA binding sites.

Alpha Lipoic Acid Promotes Development of Hematopoietic Progenitors Derived from Human Embryonic Stem Cells by Antagonizing ROS Signals

Scientists showed that alpha lipoic acid promoted production of human pluripotent stem cell derived hemogenic endothelial cells and hematopoietic stem/progenitor cells in vitro.

Glycogen Accumulation, Central Carbon Metabolism, and Aging of Hematopoietic Stem and Progenitor Cells

Inspired by recent proteomic data demonstrating the upregulation of carbon and glycogen metabolism in aging human hematopoietic stem and progenitor cells, scientists addressed whether this was caused by elevated glycolysis of the hematopoietic stem and progenitor cells on a per cell basis, or by a subpopulation that has become more glycolytic.

A 3D Engineered Scaffold for Hematopoietic Progenitor/Stem Cell Co-Culture In Vitro

To imitate the natural microenvironment, a 3D engineered scaffold for CD34+ cells co-culture was established via 3D bioprinting. The concentration of hydrogel and the ratio of two kinds of cells were optimized.

HUWE1 Cooperates with RAS Activation to Control Leukemia Cell Proliferation and Human Hematopoietic Stem Cells Differentiation Fate

To understand the role of HUWE1 in human hematopoietic stem and progenitor cells scientists constitutively expressed KRASG12V oncogene concomitantly to HUWE1 knockdown in stromal co-cultures.

GvHD-Free, Relapse-Free Survival Provides Novel Clues for Optimizing Allogeneic-HSCT for Adult T-Cell Leukemia/Lymphoma

The authors performed a nationwide retrospective study of graft-versus-host disease (GvHD)-free, relapse-free survival of patients with allogeneic hematopoietic stem cell transplantation (HSCT)-treated adult T-cell leukemia/lymphoma.

Which Lower Risk Myelodysplastic Syndromes Should Be Treated with Allogeneic Hematopoietic Stem Cell Transplantation?

The authors review prognostic factors in lower risk MDS, delineating patients with relatively poor risk who may potentially benefit from hematopoietic stem cell transplantation during the disease course.

Journals Endorse New Checklist to Clean Up Sloppy Animal Research

To address the problem of poor reporting, Percie du Sert and a team of researchers have developed a checklist of 10 critical details each animal study needs to report, such as the number of animals used, their sex, whether they were randomly allocated to a test group and control group, and whether the researchers knew which animal was in which group.
