Increased CXCL4 Expression in Hematopoietic Cells Links Inflammation and Progression of Bone Marrow Fibrosis in MPN

The authors demonstrated that the absence of hematopoietic CXCL4 ameliorated the myeloproliferative neoplasm phenotype, reduced stromal cell activation and bone marrow fibrosis and decreased the activation of pro-fibrotic pathways in megakaryocytes, inflammation in fibrosis-driving cells and JAK/STAT activation in both megakaryocytes and stromal cells in three murine primary myelofibrosis models.

Ex Vivo Editing of Human Hematopoietic Stem Cells for Erythroid Expression of Therapeutic Proteins

Scientists describe an ex vivo editing approach to achieve efficient gene targeting in human hematopoietic stem/progenitor cells and robust expression of clinically relevant proteins by the erythroid lineage.

The Periostin/Integrin-αv Axis Regulates the Size of Hematopoietic Stem Cell Pool in the Fetal Liver

Scientists showed that Itgav deletion results in increased frequency of phenotypic hematopoietic stem cells in fetal liver due to faster proliferation.

A Single Radioprotective Dose of Prostaglandin E2 Blocks Irradiation-Induced Apoptotic Signaling and Early Cycling of Hematopoietic Stem Cells

16,16-dimethyl prostaglandin E2 suppressed hematopoietic stem cell proliferation through 24 hour post ionizing radiation, which correlated with fewer DNA double-strand breaks and attenuation of apoptosis, mitochondrial compromise, oxidative stress, and senescence.

Allogeneic Hematopoietic Cell Transplantation Efficacy in Patients with Philadelphia Chromosome-Positive Acute Myeloid Leukemia in Complete Remission

Researchers retrospectively analyzed 4649 acute myeloid leukemia (AML) patients who received allogeneic hematopoietic cell transplantation and were in complete remission. Outcomes of Philadelphia chromosome-positive AML, intermediate-risk, and poor-risk AML patients were compared.

Mutation Accumulation and Developmental Lineages in Normal and Down Syndrome Human Fetal Hematopoiesis

To characterize mutation accumulation in individual stem and progenitor cells of Down syndrome and karyotypically normal fetuses, the authors clonally expanded single cells and performed whole-genome sequencing. They found a higher mutation rate in hematopoietic stem and progenitor cells during fetal development compared to the post-infant rate.

Isolation of Murine Bone Marrow by Centrifugation or Flushing for the Analysis of Hematopoietic Cells – A Comparative Study

Researchers adapted a centrifugation-based method for the isolation of murine bone marrow and compared it to the traditional flushing method. Analysis of primary hematopoietic stem cells, immune cells, and megakaryocytes revealed a comparable distribution of cellular (sub)populations.

The Impact of Donor Type on the Outcome of Pediatric Patients with Very High Risk Acute Lymphoblastic Leukemia. A Study of the ALL SCT 2003 BFM-SG and 2007-BFM-International SG

Investigators demonstrated the impact of donor type on overall results of allogeneic stem cell transplantation for very-high risk pediatric acute lymphoblastic leukemia with worse results when using mismatched donor stem cell source.

Clonal Hematopoiesis in the Inherited Bone Marrow Failure Syndromes

Scientists describe a framework by which an understanding of the processes that promote clonal hematopoiesis in inherited bone marrow failure syndromes may inform clinical surveillance strategies.

Hematopoietic Stem Cells from Pluripotent Stem Cells: Clinical Potential, Challenges, and Future Perspectives

Ex vivo hematopoietic stem cell derivation from renewable sources such as induced pluripotent stem cells offers an experimental tool for studying developmental hematopoiesis, disease modeling, and drug discovery, and yields tremendous therapeutic potential for malignant and nonmalignant hematological disorders.
