    TOP STORY

BCL6 Maintains Survival and Self-Renewal of Primary Human Acute Myeloid Leukemia Cells

Scientists explored the role of BCL6 in acute myeloid leukemia (AML). BCL6 was expressed at variable and often high levels in AML cell lines and primary AML samples.

    The
Neurotransmitter Receptor Gabbr1 Regulates Proliferation and Function of Hematopoietic Stem and Progenitor Cells

Analysis revealed that hematopoietic stem/progenitor cell numbers were significantly reduced in the bone marrow compared to wild-type littermates. Moreover, g-aminobutyric acid type B receptor subunit 1 null hematopoietic stem cells had diminished capacity to reconstitute irradiated recipients in a competitive transplantation model.

    BCL11A
Enhancer Edited Hematopoietic Stem Cells Persist in Rhesus Monkeys without Toxicity

Researchers evaluated autologous engraftment and fetal hemoglobin induction potential of erythroid-specific BCL11A enhancer edited hematopoietic stem and progenitor cells in four non-human primates.

    CSF1R
Inhibition by a Small-Molecule Inhibitor Is not Microglia Specific; Affecting Hematopoiesis and the Function of Macrophages

Scientists showed that colony-stimulating factor 1 receptor (CSF1R) inhibition by PLX5622 affected the myeloid and lymphoid compartments, caused long-term changes in bone marrow-derived macrophages by suppressing interleukin 1β, CD68, and phagocytosis but not CD208, following exposure to endotoxin, and also reduced the population of resident and interstitial macrophages of peritoneum, lung, and liver but not spleen.

    In Vivo
HSC Gene Therapy Using a Bi-Modular HDAd5/35++ Vector Cures Sickle Cell Disease in a Mouse Model

Investigators generated a HDAd5/35++ adenovirus vector containing both modules and tested it in vitro and after in vivo hematopoietic stem cell/progenitor transduction in “healthy” CD46/β-YAC mice and in a sickle cell disease mouse model.

    Pluripotent
Stem Cell Model of Shwachman-Diamond Syndrome Reveals Apoptotic Predisposition of Hemoangiogenic Progenitors

The authors established induced pluripotent stem cells from patients with Shwachman-Diamond syndrome and analyzed their in vitro hematopoietic and endothelial differentiation potentials.

Induces Autophagy in Myelodysplastic Bone Marrow Including Hematopoietic Stem/Progenitor Compartment

This study involved flowcytometric protein expression analysis of insulin growth factor receptor, PI3K‐Akt‐mTOR pathway, the autophagy related proteins, the status of antioxidative molecules SOD2 and SDF1 and apoptosis profiling in ethyl‐nitroso‐urea induced myelodysplasia.

    Human CD34+
Hematopoietic Stem Cell–Engrafted NSG Mice: Morphological and Immunophenotypic Features

Researchers histologically examined 3- to 6-month-old NSG mice, naïve or engrafted with CD34+ human hemopoietic stem cells, and employed a quantitative immunohistochemical approach to identify human and murine cell compartments, comparing the results with the fluorescent activated cell sorting data.

    CLINICAL RESEARCH

    Autologous
Hematopoietic Stem Cell Transplant in Systemic Sclerosis Is Associated with Marked Improvement in Health‐Related Quality of Life

Scientists quantified the magnitude, domains and duration of change in health‐related quality of life in patients with systemic sclerosis (SSc) treated with autologous hematopoietic stem cell transplantation (AHSCT) compared to similar SSc patients who did not undergo AHSCT.

Genetic Models of Human and Mouse Dendritic Cell Development and Function

Investigators discuss genetic models of mouse dendritic cells (DCs) development and function that have aided in correlating ontogeny with function, as well as how these findings can be translated to human DCs and their progenitors.

    All-Trans
Retinoic Acid in Non-Promyelocytic Acute Myeloid Leukemia: Driver Lesion Dependent Effects on Leukemic Stem Cells

The authors summarize the role of all-trans retinoic acid in normal hematopoiesis and in acute myeloid leukemia (AML), focusing on its impact on AML stem cells.
