Clinical and Biological Subtypes of B-Cell Lymphoma Revealed by Microenvironmental Signatures

Researchers performed a transcriptomic analysis of the microenvironment of 4,655 diffuse large B-cell lymphomas from multiple independent cohorts and described four major lymphoma microenvironment categories that associate with distinct biological aberrations and clinical behavior.

Endothelium-Derived Stromal Cells Contribute to Hematopoietic Bone Marrow Niche Formation

Scientists identified rare LNGFR+ cells in human fetal and regenerative bone marrow that co-expressed endothelial and stromal markers. This endothelial subpopulation displayed transcriptional reprogramming consistent with endothelial-to-mesenchymal transition and could generate multipotent stromal cells that reconstituted the bone marrow niche upon transplantation.

Cas9-AAV6 Gene Correction of Beta-Globin in Autologous HSCs Improves Sickle Cell Disease Erythropoiesis in Mice

Investigators used a humanized globin-cluster sickle cell disease mouse model to study Cas9-AAV6-mediated beta-globin-correction in functional hematopoietic stem cells within the context of autologous transplantation.

Mediator Subunit MED1 Is Required for E2A-PBX1–Mediated Oncogenic Transcription and Leukemic Cell Growth

The authors showed that Mediator interacted directly with E2A-PBX1 through an interaction of the MED1 subunit with an E2A activation domain. Results of MED1 depletion by CRISPR/Cas9 further indicated that MED1 was specifically required for E2A-PBX1–dependent gene activation and leukemic cell growth.

Signal-Transducing Adaptor Protein-2 Delays Recovery of B Lineage Lymphocytes during Hematopoietic Stress

Researchers exploited gene targeted and transgenic mice, and demonstrated that signal-transducing adaptor protein-2 was not required under normal, steady-state conditions.

Metabolic Sensor O‐GlcNAcylation Regulates Megakaryopoiesis and Thrombopoiesis through c‐Myc Stabilization and Integrin Perturbation

Scientists revealed for the first time that cellular O‐GlcNAcylation levels declined along the course of megakaryocyte differentiation from human‐derived hematopoietic stem and progenitor cells.

    Abstract

NAMPT/SIRT2-Mediated Inhibition of the p53-p21 Signaling Pathway Is Indispensable for Maintenance and Hematopoietic Differentiation of Human iPS Cells

Investigators demonstrated that nicotinamide phosphoribosyltransferase (NAMPT) was indispensable for the maintenance, survival, and hematopoietic differentiation of induced pluripotent stem cells.

L20, a Calothrixin B Analog, Induces Intrinsic Apoptosis on HEL Cells through ROS/γ-H2AX/p38 MAPK Pathway

Scientists investigated the anti-leukemic effect of the new Calothrixin B derivative, L20, and studied the underlying mechanisms. Cytotoxicity analysis of L20 revealed that it showed significant IC50 concentrations in HEL cells at low doses than in K562, and KG-1a.

TLR7/8 Agonist Treatment Induces an Increase in Bone Marrow Resident Dendritic Cells and Hematopoietic Progenitor Expansion and Mobilization

Chronic stimulation with Toll-like receptor (TLR)4 or TLR2 ligands resulted in an expansion of phenotypic HSCs with reduced self-renewal capacity and enhanced myeloid differentiation.

Obesity-Induced Inflammation: The Impact of the Hematopoietic Stem Cell Niche

The authors explore the role of the multicellular hematopoietic stem cell niche in hematopoiesis and inflammation, and the potential contribution of this niche to the hematopoietic response to obesity.
