    TOP STORY

Hematopoietic Stem Cell Heterogeneity Is Linked to the Initiation and Therapeutic Response of Myeloproliferative Neoplasms

Single-cell gene expression profiling with parallel mutation detection demonstrated that the megakaryocyte-primed hematopoietic stem cells subpopulation expanded significantly with enhanced potential in untreated individuals with JAK2V617F+essential thrombocythemia, driven primarily by the JAK2 mutation and elevated interferon signaling.

Facilitative Lysosomal Transport of Bile Acids Alleviates ER Stress in Mouse Hematopoietic Precursors

Scientists demonstrated that lysosomal equilibrative nucleoside transporter 3 (ENT3) transport of taurine-conjugated bile acids (TBA) facilitated TBA chemical chaperone function and alleviated endoplasmic reticulum stress in expanding mouse hematopoietic stem and progenitor cells.

The Innate Sensor ZBP1-IRF3 Axis Regulates Cell Proliferation in Multiple Myeloma

Investigators showed that Z-DNA-binding protein 1 (ZBP1) was selectively expressed in late B cell development in both human and mouse cells and it was required for optimal T-cell-dependent humoral immune responses.

    Abstract

EKLF/KLF1 Expression Defines a Unique Macrophage Subset during Mouse Erythropoiesis

Using unbiased global sequencing and directed genetic approaches focused on early mammalian development, the authors found that fetal liver macrophage exhibited a unique expression signature that differentiated them from erythroid and adult macrophage cells.

    Abstract

15-PGDH Inhibition Activates the Splenic Niche to Promote Hematopoietic Regeneration

Researchers demonstratef that the splenic microenvironment, specifically 15-PGDH high-expressing macrophages, megakaryocytes, and mast cells, regulated steady-state hematopoiesis and potentiated recovery after bone-marrow transplantation.

Overexpression of Aberrant Wnt5a and Its Effect on Acquisition of Malignant Phenotypes in Adult T-Cell Leukemia/Lymphoma (ATL) Cells

Scientists demonstrated that the overexpression of c-Myb and FoxM1 was responsible for the synergistic activation of the WNT5A promoter.

Venetoclax and Navitoclax in Combination with Chemotherapy in Patients with Relapsed or Refractory Acute Lymphoblastic Leukemia and Lymphoblastic Lymphoma

The safety and preliminary efficacy of venetoclax with low-dose navitoclax and chemotherapy was assessed in this Phase I dose escalation study in pediatric and adult patients with relapsed/refractory acute lymphoblastic leukemia or lymphoblastic lymphoma.

    Abstract

The Aurora Kinase/β-Catenin Axis Contributes to Dexamethasone Resistance in Leukemia

By combining gene expression data from cell lines and patients with acute lymphoblastic leukemia, scientists observed that the model was useful for the classification of patients.

The miR-200c/141-ZEB2-TGFβ Axis Is Aberrant in Human T-Cell Prolymphocytic Leukemia

Researchers revealed that the expression of miRNAs was aberrant and often heterogeneous in T cell prolymphocytic leukemia (T-PLL). They identified 35 miRNAs that were aberrantly expressed in T-PLL with miR-200c/141 as the most differentially expressed cluster.

    Acute Leukemia in Infants

International collaborative clinical trials have led to better understanding of the underlying molecular biology, refined risk-based stratification, and investigated the use of hematopoietic stem cell transplantation.

    Abstract

Immunotherapies Old and New: Hematopoietic Stem Cell Transplant, Chimeric Antigen Receptor T Cells, and Bispecific Antibodies for the Treatment of Relapsed/Refractory Diffuse Large B Cell Lymphoma

The authors discuss their approach to relapsed or refractory diffuse large B cell lymphoma patients and the open question of optimal sequencing of autologous transplant , CAR-T therapy, and bispecific antibodies.
