    TOP STORY
Engraftment of Human HSC in Non-Irradiated Newborn NOD-ScidIL2rγNull Mice Is Enhanced by Transgenic Expression of Membrane-Bound Human Stem Cell Factor
Scientists hypothesized that transgenic expression of human membrane-bound stem cell factor Tg(hu-mSCF) would increase levels of human hematopoietic stem cell (HSC) engraftment in non-irradiated NOD-scidIL2rγnull mice and eliminate complications associated with irradiation. [Blood] Abstract

    LABORATORY RESEARCH

ABCB6 Is Dispensable for Erythropoiesis and Specifies the New Blood Group System Langereis
Investigators identified ABCB6 as the genetic basis of the Lan blood group antigen expressed on red blood cells but also at the plasma membrane of hepatocellular carcinoma cells, and they established that ABCB6 encodes a new blood group system (Langereis, Lan). [Nat Genet] Abstract

CD27 Signaling on Chronic Myelogenous Leukemia Stem Cells Activates Wnt Target Genes and Promotes Disease Progression
In the present study, researchers investigated the possibility that the TNF receptor family member CD27 is present on leukemia stem cells and mediates effects of the immune system on chronic myelogenous leukemia. [J Clin Invest] Abstract

The Mitochondrial Transporter ABC-Me (ABCB10), a Downstream Target of GATA-1, Is Essential for Erythropoiesis In Vivo
Here scientists report for the first time that erythrocyte development in mice requires ATP binding cassette mitochondrial erythroid (ABC-me/ABCB10). [Cell Death Differ] Abstract

Alternative Splicing of Bim and Erk-Mediated BimEL Phosphorylation Are Dispensable for Hematopoietic Homeostasis In Vivo
Researchers conclude that physiological regulation of Bim relies on mechanisms independent of its alternative splicing or the Erk-dependent phosphorylation of BimEL. [Cell Death Differ] Abstract

Chromatin Modifications Induced by the AML1-ETO Fusion Protein Reversibly Silence its Genomic Targets through AML1 and Sp1 Binding Motifs
ChIP-chip analysis of human hematopoietic stem/progenitor cells transduced with the AML1-ETO fusion gene enabled scientists to identify 1168 AML1-ETO target genes, 103 of which were co-occupied by histone deacetylase 1 and had lost the hyperacetylation mark at histone H4, and 264 showed a K9 trimethylation at histone H3. [Leukemia] Abstract

Integrated Genomic Analyses Identify WEE1 as a Critical Mediator of Cell Fate and a Novel Therapeutic Target in Acute Myeloid Leukemia
Using novel molecular and computational approaches, researchers performed genome-wide short-hairpin RNA screens to identify proteins that mediate acute myeloid leukemia (AML) cell fate after cytarabine exposure; gene expression profiling of AML cells exposed to cytarabine to identify genes with induced expression in this context; and examination of existing gene expression data from primary patient samples. [Leukemia] Abstract

Disseminated Prostate Cancer Cells Can Instruct Hematopoietic Stem and Progenitor Cells to Regulate Bone Phenotype
Investigators examined if metastatic prostate cancer cells can alter regulation of normal bone formation by hematopoietic stem cells and hematopoietic progenitor cells. [Mol Cancer Res] Abstract

Ectopic Expression of the Histone Methyltransferase Ezh2 in Hematopoietic Stem Cells Causes Myeloproliferative Disease
Here scientists show that induced Ezh2 hematopoietic expression increases the number and proliferation of repopulating hematopoietic stem cells. [Nat Commun] Abstract

    CLINICAL RESEARCH

Clonal Evolution in Relapsed Acute Myeloid Leukemia Revealed by Whole-Genome Sequencing
To determine the mutational spectrum associated with relapse, researchers sequenced the primary tumor and relapse genomes from eight acute myeloid leukemia patients, and validated hundreds of somatic mutations using deep sequencing; this allowed them to define clonality and clonal evolution patterns precisely at relapse. [Nature] Abstract | Press Release

The Dynamic International Prognostic Scoring System for Myelofibrosis Predicts Outcomes After Hematopoietic Cell Transplantation
Investigators evaluated the prognostic usefulness of the Dynamic International Prognostic Scoring System in 170 patients with myelofibrosis, 12-78 (median: 51.5) years of age, who received hematopoietic cell transplantation between 1990-2009 from related (n=86) or unrelated donors (n=84). [Blood] Abstract

High-Resolution Genomic Profiling of Adult and Pediatric Core-Binding-Factor Acute Myeloid Leukemia Reveals New Recurrent Genomic Alterations
To identify cooperating lesions in core-binding factor acute myeloid leukemia, investigators performed single-nucleotide polymorphism-array analysis on 300 diagnostic and 41 relapse adult and pediatric leukemia samples. [Blood] Abstract

Unmanipulated HLA-Mismatched/Haploidentical Peripheral Blood Stem Cell Transplantation for High-Risk Hematologic Malignancies
The aim of this study was to investigate the efficacy and safety of unmanipulated haploidentical allogeneic peripheral blood stem cell transplantation for hematologic malignancies. [Transfusion] Abstract
