    TOP STORY
Proteomic Cornerstones of Hematopoietic Stem Cell Differentiation: Distinct Signatures of Multipotent Progenitors and Myeloid Committed Cells
Scientists applied an in-depth quantitative proteomic approach to analyze and compare the full proteomes of ex vivo isolated and FACS-sorted populations highly enriched for either multipotent hematopoietic stem/progenitor cells (LinnegSca-1+c-Kit+) or myeloid committed precursors (LinnegSca-1c-Kit+). [Mol Cell Proteomics] Abstract

    LABORATORY RESEARCH

The HIF Signaling Pathway in Osteoblasts Directly Modulates Erythropoiesis through the Production of EPO
Researchers show that augmented HIF signaling in osteoprogenitors results in HSC niche expansion associated with selective expansion of the erythroid lineage. [Cell] Abstract

The Histone Demethylase KDM1A Sustains the Oncogenic Potential of MLL-AF9 Leukemia Stem Cells
Using a mouse model of human MLL-AF9 leukemia, investigators identified the lysine-specific demethylase KDM1A (LSD1 or AOF2) as an essential regulator of leukemia stem cell potential. [Cancer Cell]
    Abstract | Press Release

Editing T Cell Specificity Towards Leukemia by Zinc Finger Nucleases and Lentiviral Gene Transfer
Researchers designed zinc-finger nucleases (ZFNs) that promoted the disruption of endogenous T cell receptor (TCR) beta- and alpha-chain genes. Lymphocytes treated with ZFNs lacked surface expression of CD3-TCR and expanded with the addition of interleukin-7 (IL-7) and IL-15. [Nat Med] Abstract

Regulation of Murine Normal and Stress-Induced Erythropoiesis by Desert Hedgehog (Dhh)
Scientists examined the role of Dhh in the regulation of murine erythropoiesis. [Blood] Abstract

The Polycomb Complex PRC2 Supports Aberrant Self-Renewal in a Mouse Model of MLL-AF9;NrasG12D Acute Myeloid Leukemia
Researchers used shRNA screening to identify chromatin regulators uniquely required in a mouse model of mixed lineage leukemia (MLL)-fusion acute myeloid leukemia, which revealed a role for the Polycomb repressive complex 2 (PRC2) in maintenance of this disease. [Oncogene] Abstract | Press Release

Expression of Functional P-Selectin Glycoprotein Ligand 1 on Hematopoietic Progenitors Is Developmentally Regulated
Investigators demonstrate that functional P-selectin glycoprotein ligand 1 is expressed at low levels on hematopoietic stem cells, but upregulated on the cell surface of progenitors that bear other homing molecules known to be important for thymic settling. [J Immunol] Abstract

Signal Transducer and Activator of Transcription 5b (Stat5b) Serine 193 Is a Novel Cytokine Induced Phospho-Regulatory Site that Is Constitutively Activated in Primary Hematopoietic Malignancies
Using mass spectrometry and phospho-specific antibodies, researchers identified serine 193 as a novel site of cytokine induced phosphorylation within human Stat5b. [J Biol Chem] Abstract

Protein Segregation Between Dividing Hematopoietic Progenitor Cells in the Determination of the Symmetry/Asymmetry of Cell Division
Scientists investigated how the symmetry/asymmetry of cell division in mitotic CD34+ cells can be evaluated by determining the plane of cell division and the potential distribution of proteins between daughter cells. [Stem Cells Dev] Abstract

Therapeutic Hemoglobin Levels after Gene Transfer in β-Thalassemia Mice and in Hematopoietic Cells of β-Thalassemia and Sickle Cells Disease Patients
Researchers generated lentiviral vectors carrying the human β-globin gene with and without an ankyrin insulator and compared their ability to induce hemoglobin synthesis in vitro and in thalassemic mice. [PLoS One] Abstract | Press Release

The Leukemia Associated Nuclear Corepressor ETO Homolog Genes MTG16 and MTGR1 Are Regulated Differently in Hematopoietic Cells
Investigators tried to identify structural and functional promoter elements of MTG16 and MTGR1 genes in order to find associations between their regulation and hematopoiesis. [BMC Mol Biol] Abstract

    CLINICAL RESEARCH

Results of a Pivotal Phase II Study of Brentuximab Vedotin for Patients with Relapsed or Refractory Hodgkin’s Lymphoma
In this multinational, open-label, phase II study, the efficacy and safety of brentuximab vedotin were evaluated in patients with relapsed or refractory Hodgkin’s lymphoma after autologous stem-cell transplantation. [J Clin Oncol] Abstract

First-Line Imatinib Mesylate in Patients with Newly-Diagnosed Accelerated Phase-Chronic Myeloid Leukemia
Investigators evaluated the benefit of imatinib in 42 newly-diagnosed accelerated-phase chronic myeloid leukemia patients. [Leukemia] Abstract

    INDUSTRY NEWS
Writing the Book of Cancer Knowledge
An academic-industry collaboration is releasing the first results from a new and freely available resource that marries deeply detailed cancer genome data with predictors of drug response, information that could lead to refinements in cancer clinical trials and future treatments. [The Eli and Edythe L. Broad Institute]

State Institute Awards More than $81 Million to Support Cancer Drug Discovery and Development in Texas
The Cancer Research and Prevention Institute of Texas announced funding in excess of $81 million to support cancer research and commercialization projects in Texas. [Cancer Prevention & Research Institute of Texas] Press Release

CRT and the EIF Unveil Innovative £50 Million Cancer Fund to Bridge Development Gap in the UK
Cancer Research Technology, (CRT) and the European Investment Fund (EIF) have joined together to create a £50 million investment fund to bridge the funding gap in the UK between cancer drug discovery and early development. [Cancer Research UK] Press Release
