    LABORATORY RESEARCH

RNAi Screen Identifies MAPK14 as a Druggable Suppressor of Human Hematopoietic Stem Cell Expansion
Scientists identified MAPK14/p38α as a modulator of ex vivo stem cell proliferation and showed that pharmacological inhibition of p38 dramatically enhances the stem cell activity of cultured umbilical cord blood derived hematopoietic cells. [Blood] Abstract

Extracellular Matrix Protein Tenascin-C Is Required in the Bone Marrow Microenvironment Primed for Hematopoietic Regeneration
Investigators identified Tenascin-C as a critical component of bone marrow microenvironment required for hematopoietic regeneration. [Blood] Abstract

MicroRNAs miR-125a and miR-125b Constitutively Activate the NF-κB Pathway by Targeting the Tumor Necrosis Factor Alpha-Induced Protein 3
Investigators delineated a unique epigenetic model for aberrant activation of the NF-κB pathway in cancer and provided a coherent mechanism for the role of these microRNAs in immune cell activation and hematopoiesis. [Proc Natl Acad Sci USA] Abstract

Impact of Substrate Elasticity on Human Hematopoietic Stem and Progenitor Cell Adhesion and Motility
Results indicated that matrix elasticity is an important factor in regulating the retention of hematopoietic stem cells in the endosteal niche and should be considered in attempts to propagate hematopoietic stem cells in vitro for clinical applications. [J Cell Sci] Abstract

Activin A Promotes Hematopoietic Fated Mesoderm Development through Up-Regulation of Brachyury in Human Embryonic Stem Cells
Scientists demonstrated the functional significance of Brachyury in the developmental program of hematopoietic differentiation from human embryonic stem cells and provided an in-depth understanding of the molecular cues that orchestrate step-wise development of hematopoiesis in a human system. [Stem Cells Dev] Abstract

Amino- and Carboxyl-Terminal Transactivation Domains of GATA1 Coordinate the Hematopoietic Program
Researchers demonstrated that GATA1 has two independent transactivation domains, amino (N)-terminal transactivation domain (N-TAD) and carboxyl (C)-TAD. Both N-TAD and C-TAD retained redundant as well as specific activities for proper hematopoiesis in vivo. [J Biol Chem] Full Article

Hematopoietic miR155 Deficiency Enhances Atherosclerosis and Decreases Plaque Stability in Hyperlipidemic Mice
Scientists examined the role of microRNA-155 (miR155) in atherosclerosis in vivo using bone marrow transplantation from miR155 deficient or wildtype mice to hyperlipidemic mice. [PLoS One] Full Article

Overexpression of Angiopoietin-1 Increases CD133+/c-kit+ Cells and Reduces Myocardial Apoptosis in db/db Mouse Infarcted Hearts
Researchers investigated whether angiopoietin-1 affects hematopoietic progenitor CD133+/c-kit+ cell recruitment to the infarcted myocardium thereby mediating cardiac repair in type II (db/db) diabetic mice. [PLoS One] Full Article

    CLINICAL RESEARCH

Oral Antibiotic Prophylaxis of Early Infection in Multiple Myeloma: A URCC/ECOG Randomized Phase III Study
Scientists evaluated the impact of prophylactic antibiotics on the incidence of serious bacterial infections during the first two months of treatment in patients with newly diagnosed multiple myeloma. [Leukemia] Abstract

Profiling Three-Dimensional Nuclear Telomeric Architecture of Myelodysplastic Syndrome and Acute Myeloid Leukemia Defines Patient Subgroups
Results showed distinct three-dimensional (3D) telomeric profiles specific to patients with myelodysplastic syndromes and acute myeloid leukemias that help subgroup patients based on their 3D profiles. [Clin Cancer Res] Abstract
