    TOP STORY
Spleens of Myelofibrosis Patients Contain Malignant Hematopoietic Stem Cells
Myelofibrosis (MF) is characterized by hematopoiesis occurring not only in the marrow but also in extramedullary sites such as the spleen. To study the effects of these different microenvironments on primitive malignant hematopoietic cells, researchers phenotypically and functionally characterized splenic and peripheral blood MF CD34+ cells from patients with MF. LABORATORY RESEARCH

Systems-Pharmacology Dissection of a Drug Synergy in Imatinib-Resistant Chronic Myeloid Leukemia
Integration of genome- and proteome-wide technologies enabled the elucidation of the mechanism by which a new drug synergy targets the dependency of BCR-ABLT315I chronic myeloid leukemia cells on c-Myc through nonobvious off targets. [Nat Chem Biol] Abstract

NOTCH1 Nuclear Interactome Reveals Key Regulators of Its Transcriptional Activity and Oncogenic Function
Using immunoaffinity purification, researchers identified NOTCH1 nuclear partners in T cell acute lymphoblastic leukemia cells and showed that NOTCH1 assembles a multifunctional complex containing the transcription coactivator AF4p12, the PBAF nucleosome remodeling complex, and the histone demethylases LSD1 and PHF8 acting through their demethylase activity to promote epigenetic modifications at Notch-target genes. [Mol Cell] Abstract | Graphical Abstract

Human Hematopoietic Stem/Progenitor Cells Overexpressing Smad4 Exhibit Impaired Reconstitution Potential In Vivo
Findings demonstrated that the Smad signaling circuitry negatively regulates the regeneration capacity of human hematopoietic stem/progenitor cells in vivo. [Blood] Abstract

The BAF53a Subunit of SWI/SNF-Like BAF Complexes Is Essential for Hemopoietic Stem Cell Function
Scientists provided the first evidence that the actin-related protein BAF53a, a subunit of BAF complexes preferentially expressed in long-term repopulating stem cells, is essential for adult hemopoiesis. [Blood] Abstract

Characterization of the Effects of Exercise Training on Hematopoietic Stem Cell Quantity and Function
Investigators aimed to determine whether exercise training could expand the bone marrow hematopoietic stem cell (HSC) pool, and influence various aspects of HSC function. [J Appl Physiol] Abstract

Clonal Heterogeneity in Cytogenetically Normal Acute Myeloid Leukemia Patients with NPM1 Mutations
Data demonstrated that nucleophosmin 1 (NPM1)-mutated cytogenetically normal acute myeloid leukemia originates in an early stem cell with both lymphoid and myeloid differentiation potential. [Leuk Lymphoma] Abstract

Compromised Hematopoiesis and Increased DNA Damage following Non-Lethal Ionizing Radiation of a Human Hematopoietic System Reconstituted in Immunodeficient Mice
Researchers evaluated the effects of non-lethal doses of X-ray irradiation on human hematopoietic stem and progenitor cells reconstituted in NOD/Shi-scid, IL2Rγnull immunodeficient mice. [Int J Radiat Biol] Abstract

Effect of Acute Lung Injury on VLA-4 and CXCR4 Expression in Resident and Circulating Hematopoietic Stem/Progenitor Cells
Scientists aimed to determine whether there is a relationship between pulmonary inflammation, expression of VLA-4, LFA-1, L-selectin, CXCR4, and chemotaxis in resident hematopoietic stem/progenitor cells (HSPCs), as well as the level of circulating HSPCs. [Respiration] Abstract

    CLINICAL RESEARCH

Acute Myeloid Leukemia with Translocation (8;16)(p11;p13) and MYST3-CREBBP Rearrangement Harbors a Distinctive MicroRNA Signature Targeting RET Proto-Oncogene
Researchers concluded that acute myeloid leukemia with t(8;16)(p11;p13) harbors a specific microRNA signature which is partially epigenetically regulated and which targets RET proto-oncogene. [Leukemia] Abstract

Association of Disparities in Known Minor Histocompatibility Antigens with Relapse-Free Survival and Graft-versus-Host-Disease after Allogeneic Stem Cell Transplantation
Investigators evaluated whether mismatches in a panel of seventeen minor histocompatibility antigens are associated with clinical outcome after partial T cell-depleted allogeneic stem cell transplantation. [Biol Blood Marrow Transplant] Abstract

The p53 Pathway in Hematopoiesis: Lessons from Mouse Models, Implications for Humans
Most of the understanding of p53 function in hematopoiesis is derived from genetically engineered mice. Here, researchers summarized some of these models, the various mechanisms that disrupt the regulation of p53 activity, and their relevance to human disease. [Blood] Abstract

Higher Order Orchestration of Hematopoiesis: Is Cohesin a New Player?
Scientists suggested that cohesin, which is known to mediate chromosomal cohesion between sister chromatids, may have a central role in the orchestration of hematopoiesis and may serve as a master transcriptional regulator. [Exp Hematol] Abstract
