    Stem Cell Study Opens Door to Undiscovered World of Biology
    For the first time, researchers showed that an essential biological process known as protein synthesis can be studied in adult stem cells – something scientists have long struggled to accomplish. The groundbreaking findings also demonstrate that the precise amount of protein produced by blood-forming stem cells is crucial to their function.     Circulating Hematopoietic Stem and Progenitor Cells Are Myeloid-Biased in Cancer Patients
    Researchers showed, in detailed studies of the peripheral blood from 133 untreated patients with seven different types of tumors, that the composition of circulating hematopoietic stem and progenitor cells was significantly altered in patients with solid tumors.     Efficacy against Human Acute Myeloid Leukemia and Myeloablation of Normal Hematopoiesis in a Mouse Model Using Chimeric Antigen Receptor-Modified T Cells
    Scientists showed that CD123 is a good target for acute myeloid leukemia (AML)-directed chimeric antigen receptor therapy, as its expression increases over time in vivo even in initially CD123dim populations, and that human CD123-redirected T cells eradicate primary AML in immunodeficient mice.     Donor Hematopoiesis in Mice Following Total Lymphoid Irradiation Requires Host T Regulatory Cells for Durable Engraftment
    Total lymphoid irradiation is reported to increase the frequency of regulatory CD4+CD25+FoxP3+ T cells relative to conventional T cells. Barriers to hematopoietic stem cell engraftment following this non-myeloablative conditioning were evaluated. [Blood] Abstract

    Pten Regulates Homeostasis and Inflammation-Induced Migration of Myelocytes in Zebrafish
    The expression of hematopoietic marker genes was analyzed via whole mount in situ hybridization assay in ptena and ptenb double mutant zebrafish. The embryonic myelopoiesis was characterized by living imaging and whole mount in situ immunofluorescence with confocal microscopy, as well as cell-specific chemical staining for neutrophils and macrophages.     Altered Hematopoietic Stem Cell and Osteoclast Precursor Frequency in Cathepsin K Null Mice
    Researchers investigated whether genetic ablation of CatK altered the number of hematopoietic stem cells and osteoclast precursor cells using two mouse models: CatK-/- mice and a knock-in mouse model in which the CatK gene is replaced by cre recombinase. [J Cell Biochem] Abstract

    Reduced Intensity Conditioning, Combined Transplantation of Haploidentical Hematopoietic Stem Cells and Mesenchymal Stem Cells in Patients with Severe Aplastic Anemia
    Investigators examined if transplantation of combined haploidentical hematopoietic stem cells and mesenchymal stem cells affected graft failure and graft-versus-host disease in patients with severe aplastic anemia. [PLoS One] Full Article

    Identification of Hematopoietic Progenitor Cell Donor Characteristics Predicting Successful Mobilization: Results of an Italian Multicenter Study
    Prolonged stimulation and repeated collections or failure to reach hematopoietic progenitor cell (HPC) target may occur, increasing risks for donors and recipients. Researchers carried out a retrospective multicenter study on healthy donors, to identify donor variables which may correlate with HPC mobilization. [Transfusion] Abstract

    Contributions of the Host Microenvironment to Cancer-Induced Bone Disease
    There is ever-increasing evidence for a multitude of ways in which the bone marrow microenvironment can promote disease pathogenesis, including via cancer-associated fibroblasts, the hematopoietic stem cell niche, myeloid-derived suppressor cells, and the sympathetic nervous system.
