    TOP STORY
High-Throughput Transcription Profiling Identifies Putative Epigenetic Regulators of Hematopoiesis
To elucidate the contributions of different epigenetic factors in human hematopoiesis, high-throughput cap analysis of gene expression was used to build transcription profiles of 199 epigenetic factors in a wide range of blood cells. [Blood] Abstract

    LABORATORY RESEARCH

Runx1 and Cbfβ Regulate the Development of Flt3+ Dendritic Cell Progenitors and Restrict Myeloproliferative Disorder
Scientists demonstrated that Cbfβ is essential for the development of Flt3+ macrophage-dendritic cell (DC) progenitors in the bone marrow and all DC subsets in the periphery. [Blood] Abstract

BCR-ABL-Induced Deregulation of the IL-33/ST2 Pathway in CD34+ Progenitors from Chronic Myeloid Leukemia Patients
Chronic myeloid leukemia CD34+ progenitors upregulate their cell surface expression of the IL-33-specific receptor chain ST2, proliferate and produce cytokines in response to IL-33, conversely to CD34+ cells from healthy individuals. [Cancer Res] Abstract

Critical Role of Jak2 in the Maintenance and Function of Adult Hematopoietic Stem Cells
Using a conditional Jak2 knockout allele, scientists found that Jak2 deletion results in rapid loss of hematopoietic stem/progenitors cells leading to bone marrow failure and early lethality in adult mice. [Stem Cells] Abstract

Cav-1 Deletion Impaired Hematopoietic Stem Cell Function
Researchers used Cav-1-/- mice to investigate the role of Cav-1 in hematopoietic stem cells (HSCs) function during aging. The results showed that Cav-1-/- mice displayed a decreased percentage of B cells and an increased percentage of M cells in the bone marrow and peripheral blood, and these changes were due to an increased number of HSCs. [Cell Death Dis] Full Article

Lineage-Specific Function of Engrailed-2 in the Progression of Chronic Myelogenous Leukemia to T-Cell Blast Crisis
Investigators profiled the changes in DNA methylation patterns in human samples from blast crisis (BC)-chronic myelogenous leukemia, in order to identify genes whose expression is epigenetically silenced during progression to T-cell lineage-specific BC. [Cell Cycle] Full Article

Ubiquitous Expression of MAKORIN-2 in Normal and Malignant Hematopoietic Cells and Its Growth Promoting Activity
Makorin-2 (MKRN2) is a highly conserved protein and yet its functions are largely unknown. Researchers investigated the expression levels of MKRN2 and RAF1 in normal and malignant hematopoietic cells, and leukemia cell lines. [PLoS One] Full Article

Dose Response and Clonal Variability of Lentiviral Tetracycline-Regulated Vectors in Murine Hematopoietic Cells
Researchers investigated the effect of lentiviral transduction rate on tetracycline-regulated promoter background activity, vector copy number and clonal variability as a consequence of vector integration. [Exp Hematol] Abstract

    CLINICAL RESEARCH

CD16xCD33 Bispecific Killer Cell Engager (BiKE) Activates NK Cells from MDS Patients against Primary MDS and MDSC CD33+ Targets
Researchers tested the ability of a CD16xCD33 BiKE to induce natural killer (NK) cell function from 67 myelodysplastic syndromes (MDS) patients. Compared to age-matched normal controls, CD7+ lymphocytes, NK cells, and CD16 expression were markedly decreased in MDS patients. [Blood] Abstract

The Predictive Role of Interim Positron Emission Tomography on Hodgkin Lymphoma Treatment Outcome Is Confirmed Using the 5-Point Scale Interpretation Criteria
A retrospective international multicenter study was undertaken to assess the prognostic role of interim Positron Emission Tomography performed during treatment with doxorubicin, bleomycin vinblastine and dacarbazine in patients with Hodgkin Lymphoma and the reproducibility of the Deauville five-point scale for the interpretation of interim Positron Emission Tomography scans. Relapse Assessment following Allogeneic Stem Cell Transplantation in Patients with Myelodysplastic Syndromes and Acute Myeloid Leukemia
Cytomorphology, histomorphology, and chimaerism analysis are the mainstay in any panel of post-transplant monitoring. This may be individually combined with multiparameter flow cytometry for the detection of residual cells with a leukemia phenotype and quantitative real-time polymerase chain reaction to assess gene expression, e.g., of WT1 or the residual mutation load. [Ann Hematol] Abstract
