    Genomic Landscape of CD34+ Hematopoietic Cells in Myelodysplastic Syndrome and Gene Mutation Profiles as Prognostic Markers
    Whole-genome sequencing was performed in CD34+ hematopoietic stem/progenitor cells from eight cases of refractory anemia with excess blasts, the high-risk subtype of myelodysplastic syndrome. The nucleotide substitution patterns were found similar to those reported in acute myeloid leukemia, and mutations of 96 protein-coding genes were identified. [Proc Natl Acad Sci USA] Abstract

    In Vivo Time-Lapse Imaging of Mouse Bone Marrow Reveals Differential Niche Engagement by Quiescent and Naturally Activated Hematopoietic Stem Cells
    Investigators used a natural mouse parasite, Trichinella spiralis, and multi-point intravital time-lapse confocal microscopy of mouse calvarium bone marrow to test whether hematopoietic stem cell-niche interactions may change when hematopoiesis is perturbed. [Blood] Abstract

    CBFβ and RUNX1 Are Required at Two Different Steps during the Development of Hematopoietic Stem Cells in Zebrafish
    Researchers generated and characterized two zebrafish cbfb null mutants. The cbfb-/- embryos underwent primitive hematopoiesis and developed transient erythromyeloid progenitors, but they lacked definitive hematopoiesis. [Blood] Abstract

    Antigenic-Sheltering Universal Red Blood Cells by Polydopamine-Based Cell Superficial-Engineering
    Inspired by the adhesive proteins secreted by mussels, scientists suggest a catecholic chemistry-based strategy to shelter antigenic epitopes on red blood cells (RBCs) by using polydopamine, which can guard against coagulation reaction without other negative effects on the RBCs structure, function and viability.     SIGLEC-G Deficiency Increases Susceptibility to Develop B-Cell Lymphoproliferative Disorders
    The authors studied the B-lymphocyte fate in Siglecg-/- mice over time. They demonstrated that in aging animals SIGLEC-G deficiency promotes the progressive accumulation of monoclonal B lymphocytes and increases the susceptibility to develop B-cell lymphoproliferative disorders.     Rapid Reconstitution of Functionally Active 6-sulfoLacNAc+ Dendritic Cells (slanDCs) of Donor Origin following Allogeneic Hematopoietic Stem Cell Transplant
    Investigators analyzed slanDCs, a subset of human proinflammatory dendritic cells, in hematopoietic stem cell (HSC) sources, and evaluated their one-year kinetics of reconstitution, origin and functional capacities in peripheral blood and bone marrow of patients who have undergone HSC transplant, and their presence in graft versus host disease tissue-specimens.     Identification of Hematopoietic-Specific Regulatory Elements from the CD45 Gene and Use for Lentiviral Tracking of Transplanted Cells
    Researchers report that the protein tyrosine phosphatase CD45 is an ideal candidate gene on which to base a hematopoietic reporter. CD45 regulatory elements were discovered by analyzing transcription factor chromatin occupancy and promoter nuclease sensitivity to identify minimally sufficient sequences required for expression.     Quantitation of mitochondrial apoptotic priming was used to compare susceptibility of regulatory T cells, conventional CD4 T cells and CD8 T cells to intrinsic pathway apoptosis in 57 patients after allogeneic hematopoietic stem cell transplantation and 25 healthy donors.     A Pilot Phase I Dose Finding Safety Study of the Thrombopoietin-Receptor Agonist, Eltrombopag, in Patients with Myelodysplastic Syndrome Treated with Azacitidine
    This Phase I clinical trial was designed to explore the safety and tolerability of combining eltrombopag with Azacitidine in patients with myelodysplastic syndromes.
