    Hhex Is Required at Multiple Stages of Adult Hematopoietic Stem and Progenitor Cell Differentiation
    Conventional Hhex knockouts cause embryonic lethality precluding analysis of adult hematopoiesis. Thus, scientists induced highly efficient conditional knockout (cKO) using vav-Cre transgenic mice. Hhex cKO mice were viable and born at normal litter sizes. [Stem Cells] Abstract

    ROS-Mediated Iron Overload Injures the Hematopoiesis of Bone Marrow by Damaging Hematopoietic Stem/Progenitor Cells in Mice
    The authors investigated the effects of iron overload on the hematopoietic stem and progenitor cells (HSPCs) from a mouse model. Their results showed that iron overload markedly decreased the ratio and clonogenic function of murine HSPCs by the elevation of reactive oxygen species (ROS). [Sci Rep] Full Article

    Stromal CYR61 Confers Resistance to Mitoxantrone via Spleen Tyrosine Kinase Activation in Human Acute Myeloid Leukemia
    Investigators delineated the mechanisms underlying stroma-mediated chemoresistance in acute myeloid leukemia (AML). They used two human bone marrow stromal cell lines, HS-5 and HS-27A, which are equally effective in protecting AML cells from chemotherapy-induced apoptosis in AML-stromal co-cultures.     Impact of the Immunomodulating Peptide Thymosin Alpha 1 on Multiple Myeloma and Immune Recovery after Hematopoietic Stem Cell Transplantation
    The authors observed a slight reduction in the proliferation of murine and human multiple myeloma (MM) cell lines in the presence of thymosin α1 (Tα1) in vitro. However, using two immunocompetent murine MM models, they did not observe any impact of Tα1 administration on MM development in vivo. [Cancer Immunol Immunother] Abstract

    Common Marmoset CD117-Positive Hematopoietic Cells Possess Multipotency
    As stem cell factor (SCF) is an essential molecule for hematopoietic stem cell development in mice and humans, scientists focused on CD117, the SCF receptor, and examined whether CD117-expressing cells possess the hematopoietic stem/progenitor cell characteristics of newborn marmoset-derived hematopoietic cells that can develop into T cells and B cells.     Inducible Caspase-9 Suicide Gene Controls Adverse Effects from Alloreplete T Cells after Haploidentical Stem Cell Transplantation
    To test the feasibility of a single T-cell manipulation to eliminate alloreactivity while sparing antiviral and antitumor T cells, investigators infused 12 haploidentical hematopoietic stem cell transplant patients with increasing numbers of alloreplete haploidentical T cells expressing the inducible caspase 9 suicide gene.     Researchers performed an in-depth analysis of morphologic dysplasia in 159 de novo acute myeloid leukemia cases lacking myelodysplasia-related cytogenetic abnormalities. [Mod Pathol] Abstract

    Quality of the Hematopoietic Stem Cell Graft Affects the Clinical Outcome of Allogeneic Stem Cell Transplantation

    Scientists Receive $15.7 Million to Develop Stem Cell Therapies to Treat Blood Disorders
    A consortium of scientists and transplant clinicians from the Ansary Stem Cell Institute at Weill Cornell Medical College and the Center for Cell Engineering at Memorial Sloan Kettering Cancer Center has been awarded a $15.7 million, four-year research grant from the New York State Stem Cell Science Program to translate their innovative approach to expand and manipulate hematopoietic stem cells to cure acquired and inherited blood disorders.     UB and Roswell Park Receive $1.85 Million Grant to Launch Stem Cell Research Program
    University at Buffalo (UB) and Roswell Park Cancer Institute researchers have been awarded a $1.85 million grant to create an interdisciplinary stem cell research training program. The program, called Stem Cells in Regenerative Medicine, will bring together 18 faculty members in UB’s School of Engineering and Applied Sciences and the School of Medicine and Biomedical Sciences, and Roswell Park’s Graduate Division.
