    Acceleration of the G1 Phase Transit during Cell Division Makes Human Blood Stem Cells More Powerful
    Researchers describe a new mechanism in which the length of the G1 phase of the cell cycle has a dramatic impact on the fitness of human blood stem cells. In the study, the shortened G1 phase resulted in much improved continuous production of mature blood cells from stem cells over a prolonged period of time.     Reticular Dysgenesis-Associated AK2 Protects Hematopoietic Stem and Progenitor Cell Development from Oxidative Stress.
    In zebrafish, adenylate kinase (AK)2 deficiency affected hematopoietic stem and progenitor cell development with increased oxidative stress and apoptosis. Antioxidant treatment rescued the hematopoietic phenotypes in vivo in ak2 mutant zebrafish and restored differentiation of AK2-deficient induced pluripotent stem cells into mature granulocytes.     miR-139-5p Controls Translation in Myeloid Leukemia through EIF4G2
    Researchers identified microRNA (miR)-139-5p suppression across several cytogenetically defined acute myeloid leukemia (AML) subgroups. Restoration of mir-139 expression in cell lines representing the major AML subgroups caused cell cycle arrest and apoptosis in vitro and in xenograft mouse models in vivo. [Oncogene] Abstract

    The Oncofusion Protein FUS–ERG Targets Key Hematopoietic Regulators and Modulates the All-trans Retinoic Acid Signaling Pathway in t(16;21) Acute Myeloid Leukemia
    Researchers showed that FUS–ERG acts in the context of a heptad of proteins central to proper expression of genes involved in maintaining a stem cell hematopoietic phenotype. [Oncogene] Full Article

    Inhibition of Thrombopoietin/Mpl Signaling in Adult Hematopoiesis Identifies New Candidates for Hematopoietic Stem Cell Maintenance
    The authors constructed an intracellular-truncated, signaling-deficient Mpl protein which is presented on the cell surface. The transplantation of bone marrow cells retrovirally transduced to express dnMpl into wildtype mice induced thrombocytopenia, and a progressive loss of hematopoietic stem cell.     In this multicenter retrospective study, investigators analyzed the outcome of adult patients with myelodysplastic syndrome who underwent allogeneic hematopoietic stem cell transplantation in Argentina and evaluated the prognostic factors associated with progression-free survival, overall survival, cumulative incidence of relapse, and non-relapse mortality.     Haploidentical Hematopoietic Stem Cell Transplantation: Role of NK Cells and Effect of Cytomegalovirus Infections
    NK cells have been shown to exert a key role in haploidentical hematopoietic stem cell transplantation for the therapy of high-risk leukemias.
