    CD93 Marks a Non-Quiescent Human Leukemia Stem Cell Population and Is Required for Development of MLL-Rearranged Acute Myeloid Leukemia
    Scientists showed that acute myeloid leukemias (AMLs) with genomic rearrangements of the MLL gene contain a non-quiescent leukemia stem cell (LSC) population. Although human CD34+CD38− LSCs are generally highly quiescent, the C-type lectin CD93 was expressed on a subset of actively cycling, non-quiescent AML cells enriched for LSC activity.     HDAC8 Inhibition Specifically Targets Inv(16) Acute Myeloid Leukemic Stem Cells by Restoring p53 Acetylation

    Single-Cell Transcriptomic Reconstruction Reveals Cell Cycle and Multi-Lineage Differentiation Defects in Bcl11a-Deficient Hematopoietic Stem Cells
    Investigators employed single-cell transcriptomic analysis to dissect the molecular heterogeneities in hematopoietic stem cells (HSCs). Analysis of lineage gene expression suggests that the Bcl11a-/- HSCs are constituted of two distinct myeloerythroid-restricted subpopulations. [Genome Biol] Full Article

    Hematopoietic Stem Cells Develop in the Absence of Endothelial Cadherin 5 Expression
    Utilizing time-lapse confocal imaging, parabiotic surgical pairing of zebrafish embryos, and blastula transplantation assays, researchers showed that hematopoietic stem cells emerge, migrate, engraft, and differentiate in the absence of cadherin 5 expression. [Blood] Abstract

    ATF4 Plays a Pivotal Role in the Development of Functional Hematopoietic Stem Cells in Mouse Fetal Liver
    Investigators demonstrated that deletion of activating transcription factor 4 (ATF4) significantly impaired hematopoietic development and reduced hematopoietic stem cell self-renewal in fetal liver. [Blood] Abstract

    Functional Niche Competition Between Normal Hematopoietic Stem and Progenitor Cells and Myeloid Leukemia Cells
    Scientists used the MLL-AF9 murine acute myeloid leukemia in a competitive repopulation model to investigate whether normal hematopoietic stem and progenitor cells and leukemic cells functionally compete for the same marrow niches. [Stem Cells] Abstract

    Embryonic Hematopoietic Progenitor Cells Reside in Muscle before Bone Marrow Hematopoiesis
    Researchers demonstrated that hematopoietic progenitor cell homing occurs earlier than previously reported and that hematopoietic progenitor cells reside in muscle tissue before bone marrow hematopoiesis occurs during mouse embryogenesis. [PLoS One] Full Article

    Integration of Shh and Wnt Signaling Pathways Regulating Hematopoiesis

    Amgen announced the U.S. Food and Drug Administration (FDA) has accepted for priority review the supplemental new drug application (sNDA) of Kyprolis® (carfilzomib) for injection for patients with relapsed multiple myeloma.
