    TOP STORY
Blood Stem Cell Self-Renewal Dependent on Surroundings
Using a model of the hematopoietic system, researchers have been able to precisely determine which signaling pathways play an essential role in the self-renewal of blood stem cells. A particularly decisive role in this process is the interactive communication with surrounding tissue cells in the bone marrow. PUBLICATIONS (Ranked by impact factor of the journal)
LSD1/KDM1A Promotes Hematopoietic Commitment of Hemangioblasts through Downregulation of Etv2
Scientists screened for zebrafish mutants that have defects in the hemangioblast expression of Gata1. One of the isolated mutants, it627, showed not only down-regulation of hematopoietic genes but also up-regulation of endothelial genes. They identified the gene responsible for the it627 mutant as the zebrafish homolog of Lys-specific demethylase 1 (LSD1/KDM1A). [Proc Natl Acad Sci USA] Abstract

Absence of WASp Enhances Hematopoietic and Megakaryocytic Differentiation in a Human Embryonic Stem Cell Model
Researchers used zinc finger nucleases targeted to the Wiskott-Aldrich syndrome (WAS) locus for the development of two isogenic WAS knockout (WASKO) human embryonic stem cell lines (hESCs). Upon hematopoietic differentiation, hESCs-WASKO generated increased ratios of CD34+CD45+ progenitors with altered responses to stem cell factor compared to hESCs-WT. [Mol Ther] Full Article

Identifying States along the Hematopoietic Stem Cell Differentiation Hierarchy with Single Cell Specificity via Raman Spectroscopy
Investigators demonstrated the capacity to identify discrete cells along the hematopoietic stem cells differentiation hierarchy via multivariate analysis of Raman spectra. [Anal Chem] Abstract

Human Hematopoietic Stem/Progenitor Cells Express Several Functional Sex Hormone Receptors
To better address the effects of sex hormones (SexHs) on human hematopoiesis, scientists tested for expression of receptors for pituitary SexHs, including follicle-stimulating hormone, luteinizing hormone, and prolactin, as well as the receptors for gonadal SexHs, including progesterone, estrogens, and androgen, on hematopoietic stem/progenitor cells. [J Cell Mol Med] Full Article

klf2ash317 Mutant Zebrafish Do Not Recapitulate Morpholino-Induced Vascular and Hematopoietic Phenotypes
Investigators generated a stable klf2a mutant zebrafish and characterized its cardiovascular and hematopoietic development. [PLoS One] Full Article

    CLINICAL RESEARCH

Azacitidine in Combination with Intensive Induction Chemotherapy in Older Patients with Acute Myeloid Leukemia: The AML-AZA Trial of the Study Alliance Leukemia
This randomized, controlled trial compared the efficacy of azacitidine applied before each cycle of intensive chemotherapy versus chemotherapy alone in older patients with untreated AML. [Leukemia] Abstract

Relapsed Childhood Acute Lymphoblastic Leukemia in the Nordic Countries: Prognostic Factors, Treatment and Outcome
The authors assessed outcomes following ALL relapse after common initial Nordic Society of Pediatric Haematology and Oncology protocol treatment; to validate currently used risk stratifications; and identify additional prognostic factors for overall survival. [Haematologica] Abstract

Early Application of Related SCT Might Improve Clinical Outcome in Adult T-Cell Leukemia/Lymphoma
To evaluate the impact of timing of allogenic hematopoietic SCT (allo-HSCT) for patients with adult T-cell leukemia/lymphoma (ATLL), investigators retrospectively analyzed data from patients with ATLL who received an allo-HSCT from a related donor. [Bone Marrow Transplant] Abstract
