    Repression of p53-Target Gene Bbc3/PUMA by MYSM1 Is Essential for the Survival of Hematopoietic Multipotent Progenitors and Contributes to Stem Cell Maintenance
    Scientists showed the transcriptional activation of p53 stress responses in Mysm1-/- hematopoietic stem and progenitor cells. Mechanistically, they found that the MYSM1 protein associates with p53 and colocalizes to promoters of classical p53-target genes Bbc3/PUMA (p53 upregulated modulator of apoptosis) and Cdkn1a/p21.     Stable Multilineage Xenogeneic Replacement of Definitive Hematopoiesis in Adult Zebrafish
    Investigators used a zebrafish strain lacking definitive hematopoiesis for interspecific analysis of hematopoietic cell development. Without conditioning prior to transplantation, hematopoietic progenitor cells from goldfish stably engraft in adult zebrafish homozygous for the c-mybI181N mutation. [Sci Rep] Full Article

    Co-Cultured hBMSCs and HUVECs on Human Bio-Derived Bone Scaffolds Provide Support for the Long-Term Ex Vivo Culture of HSC/HPCs
    In order to closely mimic a multi-cell state in hematopoietic stem/progenitor cells (HSC/HPCs) vascular niche, researchers co-cultured human bone marrow mesenchymal stem cells (hBMSCs) and human umbilical vein endothelial cells (HUVECs) without any cytokines as feeder cells and applied bio-derived bone from human femoral metaphyseal portion as scaffold to develop a new HSC/HPCs three-dimensional culture system.     In Vitro and In Vivo Evaluation of Cord Blood Hematopoietic Stem and Progenitor Cells Amplified with Glycosaminoglycan Mimetic
    Because glycosaminoglycans (GAGs) are known to be important components of the hematopoietic niche and to modulate growth factor effects, investigators explored the use of GAG mimetic OTR4131 to potentiate hematopoietic stem cells and progenitor cells in vitro expansion and in vivo engraftment.     Reprogramming of Human Peripheral Blood Monocytes to Erythroid Lineage by Blocking of the PU-1 Gene Expression
    To demonstrate whether PU.1 suppression induces monocyte lineage conversion into red blood cells, a combination of three PU.1-specific siRNAs was implemented to knock down PU.1 gene expression and generate the balance in favor of GATA-1 expression to induce erythroid differentiation.     ESAM Is a Novel Human Hematopoietic Stem Cell Marker Associated with a Subset of Human Leukemias
    Researchers found that endothelial cell-selective adhesion molecule (ESAM) can also be used to purify human hematopoietic stem cells from all the currently available sources (adult bone marrow, mobilized peripheral blood, and cord blood). [Exp Hematol] Abstract

    Ezh2 Regulates the Lin28/let-7 Pathway to Restrict Activation of Fetal Gene Signature in Adult Hematopoietic Stem Cells
    Scientists showed that in the absence of the polycomb-group gene Ezh2, a cohort of fetal-specific genes, including let-7 target genes, were activated in bone marrow (BM) hematopoietic stem and progenitor cells (HSPCs), leading to acquisition of fetal phenotypes by BM HSPCs, such as enhanced self-renewal activity and production of fetal-type lymphocytes.
